Research purpose:The experimental model of T2DM rats was established by high glucose and high fat diet and low dose intraperitoneal injection of streptozotocin(STZ).In the process of constructing the model,different kinds of exercise intervention were applied to explore the effects of different ways of exercise on oxidative stress and expression of Bcl-2 and Bax protein in the kidney during the formation of T2DM.Experimental method:Fifty healthy male SD rats were randomly divided into four groups after conventional adaptive feeding in a quiet environment for one week There were 10 normal control group(NC),10 diabetic model(DM),18 moderate intensity exercise group(MICT)and 12 high intensity intermittent exercise group(HIIT).NC group was fed with national standard rodent dry feed,the other three groups were fed with high sugar and high fat feed,drinking water and eating freely;After 2 weeks of adaptive exercise,all rats in NC group and DM group were free in cage for 8 weeks,while mice in MICT group and H?T group were running in different ways for 8 weeks.At the end of the 8th week,the weight and fasting blood glucose of rats in each group were measured.The rats in DM,MICT and H?T groups were given 30mg/kg intraperitoneal injection of 2%STZ solution at one time.The rats in NC group were given the same dose of citric acid buffer solution.The rats were given the overnight fasting solution at the 7th day after the injection of STZ.The weight was measured in the morning of the next day,and the blood was taken from the tail vein to measure the random blood glucose;10%chloral hydrate was used for intraperitoneal injection anesthesia,and then the plasma SOD and MDA were stored at-4? of orbital blood,and the bilateral kidneys were quickly removed and weighed,part of renal cortex was separated,and the SOD and MDA were stored in-20? refrigerator;part of renal tissue was fixed in polyformaldehyde,and the bcl-2 protein,Bax protein and renal cell apoptosis(TUNEL)were detected by immunohistochemistry;Some of them are frozen for future use.Experimental results:1.Changes of kidney weight and kidney weight index of rats in each group:Compared with DM group,the renal weight of NC group increased significantly(P<0.05),the renal weight of MICT group and H?T group increased significantly(P<0.01);compared with NC group and DM group,the renal weight index(ratio of renal weight to body weight)of MICT group and H?T group increased significantly(P<0.01);2.The changes of SOD activity,MDA content and the difference of SOD/MDA in plasma and kidney tissue of each group2.1 In plasma,the SOD activity and MDA content of MICT group were significantly lower than that of NC group(P<0.01),and that of H?T group were significantly higher than those of NC group(P<0.05);2.2 In renal tissue,SOD activity of MICT group and H?T group was significantly lower than that of NC group(P<0.05),meanwhile,MDA content of H?T group was significantly lower than that of NC group,DM group and MICT group(P<0.05),and there was no significant difference among other groups(P>0.05);In addition,the SDO/MDA value of MICT group was significantly lower than that of NC group(P<0.05),which was not statistically significant compared with DM group(P>0.05).Meanwhile,the SOD/MDA value of H?T group was significantly higher than that of DM group and MICT group(P<0.05),which was not statistically significant compared with NC group(P>0.05).3.Detection of rats in each group kidney cells by TUNEL method in each group and changes in IOD of renal cells Bcl-2 and Bax:3.1 In situ end labeling(TUNEL)was used to detect renal cells in each group,The results showed that there was no statistical significance between the groups(P>0.05).3.2 The changes of the IOD values of Bcl-2 and Bax in renal cells of rats in each group:3.2.1 Kruskal Wallis test results of IOD value of Bax are as follows:H=109.757,P<0.001,according to the test standard of ?=0.05,reject the original hy pothesis(H0),it can be considered that the difference of IOD value of Bax in e ach group is statistically significant.Bonferroni's method was used to correct the t wo comparisons after the event,the results show that:NC group,DM group and MICT group were significantly lower than H?T group(P<0.01);In addition,NC g roup and DM group were significantly higher than MICT group(P<0.05).There w as no significant difference between the other groups(P>0.05).3.2.2 Kruskal Wallis test results of IOD value of Bcl-2 are as follows:H?17.261,P?0.001,according to the test standard of ?=0.05,reject the original hypothesis(H0),it can be considered that the difference of IOD value of Bcl-2 in each group is statistically significant.Bonferroni's method was used to correct the two comparisons after the event,the results show that:NC group,DM group and H?T group were significantly higher than MICT group(P<0.05).There was no significant difference between the other groups(P>0.05).Conclusion:1.After the establishment of T2DM rat model,moderate intensity exercise and high intensity intermittent exercise did not significantly change the renal weight.2.High-intensity intermittent exercise can effectively reduce MDA of renal lipid peroxidation reaction product by increasing the ratio of SOD/MDA,so as to improve oxidative stress injury of kidney.3.Moderate exercise can inhibit renal cell apoptosis by maintaining the relativ e stability of Bcl-2 and Bax protein expression,and high-intensity intermittent exe rcise can accelerate the depolymerization of Bax-Bax homodimer by promoting the over expression of Bcl-2,thus inhibiting renal cell apoptosis.
