Objective:To investigate the expression of rs1126616,rs11728697,rs4754 and rs11730582 SNPs of secreted phosphoprotein 1(SPP1)genes in cervical cancer and the control group,and to further analyze the relationship between the polymorphism of rs11730582,rs11728697,rs4754 and rs1126616 genes and the genetic susceptibility of cervical cancer.To study the distribution of secreted phosphate protein 1(SPP1)in the serum of cervical cancer patients and healthy controls,and to analyze its relationship with the clinicopathological features of cervical cancer patients.Methods:Use case-control study method,using multiple SNPS classification technology(im LDRTM)to detect 379 cases of cervical cancer and 350 healthy women SPP1rs11730582 site,rs11728697 loci,rs4754 site and genotyping rs1126616 loci,each point total number of genotypes and alleles,frequency can be calculated directly,and the comparison between different groups by chi-square(χ2),adjustment for age in logistics regression analysis to calculate the OR and 95% CI,The hardy-weinberg balance was used to test the rexpresentativeness of the population.Meanwhile,the serum SPP1 content of 255 patients with cervical cancer and 268 healthy controls was determined by double-antibody sandwich ELISA,and the distribution of SPP1 content in the clinicopathological features of cervical cancer was analyzed.Use of relevant statistical methods for each laboratory test data.Statiastical significance was assumed at the P﹤0.05 level.Results:(1)there was a statistically significant difference in the distribution of CC,CT and TT genotypes at rs11728697 between the two groups(P < 0.05).The frequency of CT genotypes in the cervical cancer group was higher than that in the control group,and carryingCT genotypes would increase the risk of cervical cancer(OR=1.401,95%ci: 1.023~1.918,P=0.035).(2)the distribution of CC,CT and TT genotypes at rs11730582 was statistically significant between the two groups(P < 0.05).The frequency of CT genotypes in the cervical cancer group was higher than that in the control group,and carrying CT genotypes would increase the risk of cervical cancer(OR=2.157,95%ci: 1.295~3.594,P=0.003).(3)there is a weak linkage imbalance between partial loci of rs1126616,rs11728697,rs4754 and rs11730582.(4)there were 11 haplotypes at four sites of rs1126616,rs11728697,rs4754 and rs11730582,among which TCCT,TCTC and CTCT haplotypes were the three main haplotypes in the cervical cancer group and the control group.ELISA results showed that SPP1 was positively expressed in the serum of both cervical cancer patients and healthy controls,but its content in the serum of cervical cancer patients was significantly higher than that of healthy controls,and the difference was statistically significant(P < 0.05).The expression of SPP1 was not correlated with the pathological type of cervical cancer,but positively correlated with the clinical stage and differentiation degree of cervical cancer.In other words,the higher the stage and the lower the differentiation degree of cervical cancer were,the higher the serum content of SPP1 was.
