Based on the idea of processing integration in the original processing place,this paper mainly studied the processing and drying method of traditional Chinese medicine Polyporusumbellatus?Pers.?Fries.The control model is established to monitor the machining process.At the same time,the drying mechanism of different drying methods was studied.The effect of drying mode on the quality of crude drugs was further expounded.In this paper,the dry processing of polyporus in different ways was studied.This experiment includes the following aspects.Chapter 1 The establishment of quality evaluation index of polyporusIn the 2015 edition of Chinese pharmacopoeia only ergosterol as used to as the quality evaluation index of Chinese polygonatum.Combined with the literature,it is found that in recent years,there is little research on the establishment of quality control index of polyporus.In the study,a method for the determination of ergosterol,ergosterone and polysaccharide was established based on the optimization of the method.Polyporus is a traditional diuretic,in which the steroid components ergosterol,ergosterone play the main role.The UPLC method was used to determine the contents of two components simultaneously at different wavelengths.It was found through literature review in the early stage,there are many methods to determine the content of polyconfections,the affinity of the same polysaccharide to different methods are also varies.The methods of determination of polyporuspolyccharide by anthranone sulfate and phenol sulduric acid were compared.Optimum determination method and technology craft.The original reflux extraction method of PPS was improved,and the extraction process of PPS was optimized by ultrasonic assisted extraction combined with response surface method at the same time.On the basis of single factor,ultrasonic extraction time,ultrasonic temperature and liquid-material ratio were optimized.Chapter 2 The drying process of Chinese traditional medicine polyporus was optimized based on muti-index comprehensive scoring methodBy comparing the drying method?50?,60?,70?,80?,respectively?,microwave drying method and freeze drying method with traditional natural drying method,the effects of different drying methods and different drying temperature on the contents of main chemical components in polyporus were analyzed.By comparing the changes of chemical constitution in polyporus processed by different drying methods,the method of drying and processing was selected based on the content of each component.At the same time,the antioxidant activity of PPS was measured.The scavenging effects of extracted and purified PPS on hydroxyl radical,DPPH radical and superoxide radical were investigate.Based on the hierarchical weight analytic hierarchy process?AHP?,according to the nature of problem and the ultimate goal to be achieved,the different levels are gathered together through the subordination relationship,and the measured indicators are quantitatively analyzed to overcome the subjective evaluation..Comprehensive optimization of the best processing scheme.Chapter 3 Study on the changes of color and taste of polyporus with electronic nose and electronic eyeIn this experiment,the electronic nose and electronic eye technology were used to measure the processed polyporus,to study the changes of color and odor of processed polyporus under different drying processes,to use principal component analysis and discriminant factor analysis to process date,and to establish a quality control analysis model.Chapter 4 Effects of different drying processes on the physical properties and structure of Chinese herbal medicinesStaring from the microscopic characterization and microscopic structure of medicine materials,the relationship between drying method and structure change and physical property characterization was explored.To investigate the rehydration rate,hardness and dissolution effect of prepared slices,and to observe the structure of different samples by scanning electron microscopy under microscopic conditions.The Image J software was used to measure the porn circumference and porn area in different dried chine rinds.The concept of fractal dimension under drying condition was induced and the fractal characteristics of drying 0.5 1%,0.95%and 1.88%,respectively,and the RSD values odergosterone accuracy,stability and resproducibility are 0.27%,1.04%and 0.64%,respectively.The established method is stable and reliable,can be used to evaluate the quality of polyporus.The result of snthranone sulfate method is more stable and reliable with high repeatability.This method is suitable for the determination of polysaccharide in polyporus.The determination condition of anthranone sulfate method is as follow,1m L polysaccharide solution is added to 2.5 m L anthranone sulfate of 0.02 mg·m L-1,and the reaction was carried out under water bath at 70?for 5 min.The optimal process is determined as the ultrasonic time of 60 min,the ratio of liquid to material is 100:1,and the ultrasonic temperature is 60?.Verification experiments confirm that the process is stable.The contents of ergosterol and ergosterone in 18 batches of polyporus were determined.Ergosterol content was 1.798mg·g-1-3.969 mg·g-1.The content of ergosterone was 0.041 mg·g-1-0.095 mg·g-1.which was in line with the requirements of Chinese pharmacopoeia.The contents of.polysaccharide in 18batches of polyporus were determined.The polysaccharide content was 19.569 mg·g-1-24.230mg·g-1.Different drying temperature and different drying processing methods have no significant effect on the content of steroid components,but have significant effect on the polysaccharide components of polyporus.80?drying and microwave drying have significant effects on the retention of polysaccharide components,while the natural drying method,which is widely used at present,have poor effects on the retention of polysaccharide components in PPS.At the same time,adopting the processing technology of processing integration in the producing area of polyporus has significant advantages,which can avoid the process of drying and softening the herbs once again.The effect of different drying condition on the antioxidant activity of PPS were compared,the result showed that the antioxidant activity was the strongest when the decoction pieces were dried during 50-60?.Drying at low temperature and high temperature can inhibit the antioxidant activity of PPS.In the study,the contents of ergosterol,polysaccharide,ergosterone,antioxidant activity and extract in polyporus were evaluated by comprehensive scoring.On the basis of the comprehensive score,it was determined that when drying at 80?,the comprehensive effect of each index in the medicinal materials reached he best.Determine the origin of polyporus processing integration drying process.To promote the establishment of processing and processing standards in producing areas of polyporus,to ensure the quality of herbs and slice to improve the rationality of clinical application,and to provide the research.And there is no significant difference in the smell of polyporus under different techniques.Under different drying condition,there is obvious difference in the color of polyporus.Through statistical quality control analysis?SQC?,distinguish between processed products,a drying model of 80?is established to provide ideas for further promoting the application of electronic nose and electronic eye in the control of processing process,quality identification control and reject of qualified products.The result shows that the increase of growth years,the porn structure is more obvious,the rehydration effect is stronger and the hardness decreased.By measuring different batches of polyporus produced by medicinal materials from different places and manufactures,the experimental results further indicated that the rehydration rate of polyporus is positively correlated with the size of porn structure,and inversely correlated with the hardness of polyporus.Different drying method has effects on the structure of the slices.By calculating the average pore area and the average pore circumference,the results shows that the logarithm of the area and the logarithm of the circumference have a linear relationship,and?r=0.955?,and the fractal dimension of different drying methods is different.With the increase of drying temperature,the fractal dimension decrease gradually.Combined with the dimension of polysaccharide components in decoction pieces,it is found that the smaller the fractal dimension is,the shorter the time needed for the dissolution of decoction to reach the maximum dissolution effect,and the better the dissolution effect of components is,suggesting that that increase the drying temperature within a certain range is conductive to improving the dissolution rate components.
