ObjectiveResearch on acupuncture potential suitable condition.Exploring the network regulation of acupuncture analgesia and anti-inflammatory.Methods10 Wistar rats were injected with 0.1 m L of complete Freund's adjuvant into the right hind foot sole to induce inflammation.The 10 rats with successful modeling were randomly divided into complete Freund adjuvant model group and complete Freund adjuvant model + electroacupuncture group,5 rats in each group.The other 12 rats were randomly divided into control group and electroacupuncture group,6 rats in each group.Rats in the model electroacupuncture group and normal electroacupuncture group were stimulated with electroacupuncture at ST36 and GB30 points for 30 minutes each time and once every other day.After day 27,rats in each group took blood from the abdominal aorta,and centrifuged to extract serum.Serum exosomes were extracted using the Exo Quick kit method,and label free quantitative proteomics technology was used to identify the types and quantities of serum exosome protein components.Screening for significantly different proteins with acupuncture by the criterion of change multiple greater than 1.5 and p <0.05.Using significant differential proteins screened from the results of the proteomics as the source data,and find diseases associated with significantly different proteins in the MALACARD database and build significant protein-disease network.Data were analyzed by degree,weighting degree,and Page Rank to screen for diseases with a high degree of correlation with significantly different proteins and predict potential suitable conditions for electroacupuncture.Finds the interaction proteins,signal pathways,Molecular function and significant differences in protein sources in cells and tissues.Build a significant difference of protein-protein interaction network,the significant difference of protein-signal through network,and network significantly different protein-function significantly different protein-secretory cell organization network.Data were analyzed by running algorithms such as penetration,weighting,and Page Rank,and key interaction proteins,signaling pathways,molecular functions,and secretory cell tissues were screened.Results1.Potential advantages of acupuncture includes 32 types of tumors,15 types of immune system diseases,10 types of blood system diseases,6 types of infectious diseases,5 types of kidneys,5 types of cardiovascular diseases,4 types of metabolic diseases,4 types of motor system diseases,2 types of digestive diseases,2 types of respiratory diseases,2 types of skin diseases,2 types of neurological diseases,1 types of liver diseases,1 types of facial diseases,and 12 other diseases.2.It was found that the molecular function of significantly different proteins involves the following aspects:bioactive substance binding,bioactive substance transport,immune regulation,signal transduction,and bioactive substance metabolism.The interaction protein function involves the following aspects:immune regulation,lipid metabolism,energy metabolism,signaling,hemoglobin composition,hemolytic coagulation,and so on.Signaling pathways such as Fox O,Ras,complements and clotting cascades,HIF-1,glycoenzyme/sugar isogenesis,and PPAR may be key signaling pathways for the network regulation of acupuncture,many of which are associated with immunomodulation,energy metabolism,lipid metabolism,etc.Significantly different proteins in acupuncture can come from various systems such as digestive system,reproductive system,respiratory system,endocrine system,circulatory system,immune system,exercise system,nervous system,urinary system,skin system,etc.Conclusions1.Acupuncture has a wide range of potential adaptation disorders,involving multiple system diseases,of which immune system diseases,tumors,and metabolic diseases may be important potential predominant diseases for acupuncture.2.Acupuncture analgesic and anti-inflammatory effects have the characteristics of network regulation,and can play an overall role from organs,tissues,cells,proteins,signaling pathways and other multi-level,multi-dimensional,multi-channel.
