Objective:By establishing a rabbit VX2 liver tumor model and using the synthesis of polidocanol and iohexol as embolization materials,the feasibility,safety and efficacy of the synthesis of polidocanol and iohexol as new embolization materials for liver cancer were explored.Materials and methods:The VX2 liver tumor model was established in 20 rabbits from New Zealand.After 14 days,the tumor size was measured by MR scanning.The rabbits were divided into two groups,A and B,by random number method.The tumor was treated with hepatic artery embolization.Group A consisted of the synthesis of polidocanol and iohexol,and group B consisted of sterilized water for injection.Serum ALT,AST,Cr,BUN were detected to evaluate changes in liver and kidney function.After 7 days of embolization,MR scanning was performed again to evaluate the therapeutic effect of tumor,and the tumor growth rate was calculated.The experimental rabbits were then sacrificed and the appearance of liver,heart,lung,spleen and kidney was observed.Two independent sample t test was used for comparison among groups,the analysis of variance?anova?of repeated measurement design was used for intra-group comparison.Results:All the 20 experimental rabbits were implanted with liver tumors using open surgery,of which 14 experimental rabbits were successfully implanted with hepatic artery embolization,with an overall success rate of 70%.Changes of liver and kidney function:on the 1st,3rd and7th day after embolization,ALT level of experimental rabbits in group A was increased compared with that in group B?all P<0.05?.On the 1st and 3rd day after embolization,the AST level of experimental rabbits in group A was higher than that in group B?all P<0.05?.On the7th day after embolization,there was no significant difference in AST level between group A and group B?P>0.05?.There was no statistically significant difference in Cr and BUN levels on day1,day 3 and day 7 after embolization in rabbits in the two groups?all P>0.05?.There were statistically significant differences in ALT levels of experimental rabbits in group A before and after surgery on day 1,day 3 and day 7?all P<0.05?.There were statistically significant differences in AST levels before surgery,on day 1 and day 3 after surgery?all P<0.05?,and no statistically significant differences between AST levels before surgery and on day 7 after surgery?P>0.05?.There was no statistically significant difference in Cr and BUN levels in rabbits in group A before and after surgery on day 1,day 3 and day 7?all P>0.05?.There were no statistically significant differences in ALT,AST,Cr and BUN levels in the experimental rabbits of group B before and after surgery on day 1,day 3 and day 7?all P>0.05?.Before embolization,the tumor volumes of each group were:group A?975.2±745.9?mm and group B?1060.0±622.9?mm,respectively.There was no statistically significant difference between the two groups?t=0.231,P=0.821?.On the 7th day after embolization,the tumor volumes of the two groups were:group A?2370.1±1561.5?mm and group B?5213.0±2434.1?mm,respectively.The difference between the two groups was statistically significant?t=2.601,P=0.023?.The tumor growth rates of the two groups were:?160.4±53.7?%in group A and?489.8±348.4?%in group B,respectively.The difference was statistically significant?t=2.473,P=0.046?.The tumor growth rate of group A was lower than that of group B.In group A,both the tumor tissue and the surrounding liver tissue presented coagulative necrosis,and the boundary was not clear.In group B,the tumor tissue and the normal liver tissue were decomposed clearly,and the tumor cell atypia was increased and the tumor tissue was active.Pathological examination of heart,lung,spleen and kidney:the core,lung,spleen and kidney in the two groups of experimental rabbits were in normal general shape,with no obvious edema and necrosis,and no damage was observed in sections under the light microscope.Conclusion:The synthesis of polidocanol and iohexol is safe and effective as a new embolization material for HCC and has potential value as a new embolization material for TACE treatment of HCC.
