Objective:To explore the damage degree of vascular ethoxylated alcohol and iohexol in the liver and liver tissues of rabbits under the guidance of DSA,and to investigate the safety of ethoxylated alcohol and iohexol as arterial embolization agents.Effectiveness.Methods:Eighteen New Zealand rabbits were randomly divided into 3 groups(immediate group,1-day group and 3-day group),6 rabbits in each group,and all the experimental rabbits underwent 3.0T MR scanning at 1 day before surgery.The model of hepatic artery embolism was established by femoral artery puncture in experimental rabbits.Selecting the left or right hepatic artery and 0.4ml of ethoxylated hard alcohol and iodixanol was slowly injected into the hepatic artery under angiography mode.Immediately after the injection,DSA images were taken at intervals of 10 minutes.All rabbits underwent 3.0T MR scan immediately after hepatic artery catheterization and embolization,1 day and 3 days.According to the different groups,the experimental rabbits were sacrificed at the corresponding time.After the injection of the drug,the liver specimens were fixed in formalin solution to observe the change of the contrast image,and the liver appearance was observed.The liver tissue of the injected drug was taken for HE staining and silver staining.dyeing.Results:1.Establishment of hepatic artery embolism model: 18 experimental rabbits,16 of them were SUPER-SELECTED successfully and injected with drugs,the success rate was 88.8%.2.DSA angiography: Compared with preoperative angiography,there was no obvious filling defect and occlusion of vascular stenosis in immediate post-operative angiography and 10 minutes post-operative angiography.3.MR manifestations: Normal hepatic parenchyma showed moderate signal on TIWI and T2 WI.Immediate group: There was no change.1-day group: The hepatic tissue of injected drug showed slightly high signal intensity on TIWI and T2 WI.3-day group: The hepatic tissue of injected drug showed slightly high signal intensity on TIWI and T2 WI.4.Gross pathological manifestations: Immediate group: There was no difference between normal liver tissue and drug-damaged tissue,and the boundary was not clear.1-day group: The liver tissue of the injected drug became flaky white,and the tissue destroyed by the drug was softer and clearer than that of the normal liver tissue.3-day group: The liver tissue of the injected drug was lamellar brown with hard texture,clear boundary and caseous necrosis.The surrounding liver tissue was lighter in color and the liver tissue granules were thicker than the normal liver tissue.5.Light microscopic findings: Immediate group: HE staining showed normal morphology of hepatocytes and structure of hepatic lobules,edema of hepatocytes,normal structure of hepatic sinuses,and no obvious abnormalities of interlobular arteries and veins.There was no obvious abnormality in argyrophilic staining.1-day group: HE staining showed that hepatocyte morphology and lobular structure existed,sinusoidal dilatation,endothelial damage,hepatocyte nucleus blurred,cytoplasm reduced,early coagulative necrosis,vascular endothelial discontinuity was observed in interlobular artery and interlobular vein,and thrombosis was observed in them.The periphery of necrosis was marked by hyperemia and edema,accompanied by more neutrophil infiltration,and liver reticular fibrous structure was destroyed by argyrophilic staining.3-day group: HE staining showed homogeneous red staining of hepatocytes,disappearance of nuclei,and complete coagulation necrosis of hepatic lobules.A large number of neutrophils infiltrated around the necrosis,and hepatic reticular fibrous structure was destroyed by argyrophilic staining.Conclusion:Ethoxylated alcohol and iohexol are an injectable embolic agent,which areconvenient and visible,causing damage to vascular endothelium,hepatocytes and hepatic sinusoids,and then induces intravascular thrombosis.They have potential applications of materials as a hepatic artery embolism.
