Objective:To investigate the effect of eumame extract on the expression of Tamm-Horsfall protein?THP?in the renal stone model of rats induced by nanobacteria.The possibility of preventing the formation of kidney stones in rats was discussed,which provided a new clinical idea for the prevention and treatment of kidney stones.Methods:Kidney stones?excluding infections,endocrine diseases,multi-system stones and other diseases?from 10 patients who underwent urologic surgery in our hospital were collected.The kidney stones were demineralized,neutralized and washed,then crushed,centrifuged,filtered and cultured.Finally,the nanobacteria from kidney stones were identified by PCR.45 SD male rats were randomly divided into blank group,control group and experimental group,15each.Rats in the control group and the experimental group were injected with 1m L of nanobacteria suspension via tail vein to prepare the rat kidney stone model,and rats in the blank group were injected with the same amount of normal saline via tail vein.Rats were given 1m L eumame extract on the second day,and the blank group and control group were given the same amount of normal saline.After 4 weeks of continuous intervention,urine was collected in a metabolic cage for 24 hours.Urine p H,Ca2+,citric acid?CA?and oxalic acid?Oxa?were detected.The expression of THP in urine was determined by ELISA.Kidney tissues were stained with HE,and pathological changes of the tissues and the crystal morphology and amount of calcium salt in the kidney tissues were observed.Results:Compared with the blank group,control group in the kidney tissue calcium salt crystallization integral?3.80±0.77?significantly increased?P<0.05?,urine p H value?5.75±0.21?and CA levels?4.17±1.49?mol/L?decreased significantly?P<0.05?,Ca2+?3.11±0.16mmol/L?,level of Oxa?210.14±20.97?mol/L?and THP?61.63±3.83mg/L?significantly increased expression?P<0.05?,HE staining showed that renal tubular serious injury,atrophy,renal tubules surrounding tissue inflammatory cells infiltrating.Crystal formation of calcium oxalate was observed in the interstitium of the kidney.Compared with control group,experimental group in the kidney tissue calcium salt crystallization integral?2.60±0.51?decreased significantly?P<0.05?,urine p H value?6.66±0.13?and CA levels?37.58±3.99?mol/L?significantly increased?P<0.05?,Ca2+?2.73±0.18mmol/L?,the level of Oxa?175.84±12.87?mol/L?and THP?56.27±3.13mg/L?expression decreased significantly?P<0.05?,HE staining showed that renal tubular atrophy of focal lesions,and focal inflammatory cell infiltration,renal small tube can be found in asmall amount of brown crystal deposition.Conclusion:Dark plum extract may inhibit the formation of calcium oxalate crystals in kidney tissues by reducing the expression of THP and self-polymerization in rats with nanobacteria-induced kidney stones.
