Objective:Observation on the effectes of Astragalus and Dark Plum Extracts on Insulin Resistance Caused by High Fat Fed and Dexamethasone in Rats.Methods:40 male wistar rats, four to six-week-old, body weight 240±20g, were randomly divided into two groups: normal control group(n=5)and high-fat diet and dexamethasone group(n=35).Choose the rats which have higher fasting blood glucose and abnormal glucose tolerance as successful IR rats. Then all the mice were divided into four groups: control group,insulin resistance model group, extracts of astragalus and dark plum group and shenqijiangtang gradule granule group. Blood glucose, glucose tolerance, blood lipid and serum insulin were measured by different methods. Observe the liver tissue pathological changes by HE staining; Sq RT-PCR assess the expression of IRm RNA and GLUT4 m RNA.Results:(1) Compared with the normal diet group rats, FBG of mice in model group was significantly higher(p<0.05), and the tolerance of glucose in the model group increased significantly(p<0.05). After received the extracts of astragalus and dark plum, FBG and Homa-IR decreased significantly.(2) TC, FFA of mice in model group were higher than that of mice in control group(p<0.05). After received the extracts of astragalus and dark plum, TCand FFA of mice decreased significantly(p<0.05).(3) Liver tissue pathological of mice in model group changes as fat appearance,cavitation fat cells emerged, extracts of astragalus and dark plum can reduce the damage.(4) Quantity of In R m RNA expression of liver tissue and GULT4 of skeletal muscle of model group rats was obviously lower than normal group(p < 0.05), after drug intervention,in two drug intervention group, quantity of Ins R m RNA expression of rats increased, mice in extracts of astragalus and dark plum group increased more significantly(p < 0.05).Conclusion:Extracts of astragalus and dark plum can effectively reduce the blood sugar of insulinresistance model rats and correct the disorder of lipid metabolism, its mechanism may be related to the increase the IRm RNA and GLUT4 m RNA in liver and skeletal.
