Objectives :To analyze the expression and significance of interleukin(IL)25 in patients with non-alcoholic fatty liver disease(NAFLD)liver fibrosis.Methods:We enrolled 36 patients with NAFLD and 20 patients in the control group who met the inclusion criteria from June 2018 to December 2019.According to liver stiffness,patients with NAFLD were divided into mild liver fibrosis group(F1)20 cases and significant liver fibrosis group(F ? 2)16 cases.The serum IL-25 level of patients in each group were measured.Patients with abnormal transaminase were selected from the NAFLD group for liver biopsy.In the control group,mild liver fibrosis group and significant liver fibrosis group,each group selected 6patients to obtain liver tissues,and real-time fluorescent quantitative PCR was used to detect the liver tissues IL-25,COLLAGEN I,? mooth muscle actin(?-SMA),macrophage mannose receptor(CD206),glutamine transaminase 2(TGM2)mRNA expression levels.Immunohistochemistry or immunofluorescence was used to detect the protein expression levels of IL-25,a-SMA,CD206 and TGM2.Results : There was no significant difference in serum IL-25 between the control group(26.51±5.84pg/ml),NAFLD with mild liver fibrosis group(24.18±4.80pg/ml)and NAFLD with significant liver fibrosis group(24.32±6.02pg/ml)(F = 1.41,P = 0.254).The hepatic mRNA expression and protein levels of IL-25 of significant fibrosis group were significantly lower than those in the control group(P values 0.008 and,< 0.001 respectively)and mild fibrosis group(P values 0.024 and,0.002 respectively).The hepatic mRNA expression levels of CD206 and TGM2 of significant fibrosis group were significantly lower than those in the control group(P values 0.003 and,< 0.001respectively)and mild fibrosis group(P values 0.004 and 0.045,respectively).The hepatic M2 macrophages were significantly more than those in the control group and mild liver fibrosis group(P values 0.001 and < 0.001,respectively).Conclusions: With the emergence and progress of NAFLD liver fibrosis,thedecline in expression of IL-25 in liver tissues is accompanied by a decrease in activation of M2 macrophages.
