ObjectiveTo investigate the effect of interleukin-18in nonalcoholic fatty liverrats with electroacupuncture therapy and discuss the therapeutic mechanismof acupuncture to nonalcoholic fatty liver.Methods44adult male SD rats were randomized into two groups:33in model group and11in control group. Rats in model group had be givenhigh sugar and high fat diets for5weeks into NAFLD model. After the successful model,they were randomly divided into model group(n=10),electro acupuncture group(n=10) and non acupuncture group(n=10).Rats in electroacupuncture group were acupunctured "Fenglong"(ST40),"Zusanli"(ST36),"Taichong"(LR3) and "Sanyinjiao"(SP6). Rsts innon acupuncture group were acupunctured four points randomly in one third section of their tails. After4weeks of the treatment, the fasting blood glucoses of rats were detected by enzymic method and thefasting plasma insulins of rats were detected by radioimmunoassay. Fas ting insulin resistance indexs were calculated and the pathological morphology of rats livers were observed.The level of interleukin-18in the serums and liver homogenate tissues from rats was tested by double antibody sandwich enzyme-linked immunosorbent serologic assay.ResultsAfter four weeks, compared with control group, the levels of FBS、FINS、FIRI and IL-18in the serums and liver homogenate tissues in modelgroup increased significantly (P<0.01). There were various degrees ofhepatic steatosis in model group.Compared with model group, the levels ofFBS、FINS、FIRI and IL-18in EA group decreased significantly (P<0.05).Hepatic steatosis in EA group was relieved.No significant difference wasfound in model group and NEA group about above indexes(P>0.05).Therewas no obvious improvement in NEA group.ConclusionsElectroacupuncture stimulating special points can treat rats withNAFLD whose mechanism is through declining IL-18in the serums andliver homogenate tissues,reducing lipids in hepatic tissues and blockinghepatic fatty degeneration and inflammation-injury.
