ObjectiveInvestigate the therapeutic effects and side effects of thyroxine on fat-inducednon-alcoholic fatty liver.Methods1、Health4weeks84SD male rats, repurchase all the animals and feed one week adaptively,if there are no adverse reactions, eat and drink normally, the animal can be included in theexperiment. The SD rats were divided into8groups randomly: normal group, high-fatgroup, low-dose thyroxine prevention group (4mg/kg.d) group, high-dose thyroxinePrevention (8mg/kg.d) group, low-dose thyroxine treatment (8mg/kg.d) group, high dosethyroxine treatment (4mg/kg.d) group, gemfibrozil group, saline group, in addition to thenormal group, all the SD rats were fed with high-fat diet and tap water, all drugs weredissolved in saline, the intervention group were weighed once a week, according toweight change the amount of drug and gather tail blood at the0,4,8week.Gatherabdominal aortic blood at the12week, slaughter the SD rats, take the liver.2、Detect the indicators of all the rats in each group: observe general condition of each group,weighed weekly,after the rats were sacrificed,gather the blood and liver tissue, observethe general appearance of the liver and calculate the liver index (liver wet weight/weightx100%), and detect the serum AST、ALT、TG、TC、 CK、LDH levels; each group of liveris stained by tissue HE staining and Masson staining of collagen,oberserve thepathological changes in rat liver tissue;The rats serum thyroid hormone(T3, T4,TSH)changes were detected at each time point before and after treatment by using ELISAmethod.Results(1) The effect of thyroxine on rat body weight, liver index: the weight of the rats were increased at the end of modeling, The weight of the rats from high-fat group and thesaline group were the fastest growthing groups when compared with the normal group (P<0.05).Compared with age-fat group,The weight of the rats from thyroxine preventiongroup and gemfibrozil group were grouth slowly.and its liver index was significantlylower than the high-fat group (P<0.05); weight and liver index have no statisticallysignificance between other groups (P>0.05).(2) The effect of thyroxine on rat pathology: According to HE staining and Masson stainingof collagen, the high-fat group, saline group, thyroxine group, gemfibrozil rats pathologyscores and analysis collagen area were significantly higher than the control group (P<0.01);thyroid prevention group and gemfibrozil group pathology scores and analysiscollagen area lower than the high-fat group and the saline group (P <0.05); thyroxinetreatment group pathology scores and analysis collagen area was not statisticallysignificant (P>0.05); the high-fat group and the saline group was not statisticallysignificant (P>0.05).(3) The effects of thyroid hormone on serum indexes of rats: Compared with the normalgroup, high-fat group and the intervention group rat serum ALT, AST, TC, TG levelswere significantly higher (P <0.05); intervention group in prevention group, highergemfibrozil group serum lipid group ALT, AST, TC, TG levels were significantlyimproved (P <0.05), no significant difference between the groups. Each group of ratsserum enzyme CK, LDH were within normal range.(4) The serum thyroid hormone levels in rats of every group: Compared with the same periodin the normal group, the serum T3,T4levels were slightly lower and the TSH wereincreased in high-fat group;The serum T3,T4levels were slightly lower and the TSH wereincreased at the12week in saline group; Compared with the same period in high-fatgroup, the serum T3、T4were increased at8week,12week in thyroxine preventiongroup and gemfibrozil group (P <0.05), the thyroxine treatment groups was notstatistically significant at the12week (P>0.05).Conclusion1、Thyroid hormone can reduce fat-induced lipid levels in rats with nonalcoholic fatty liver, improve liver function and liver pathological changes, has a protective effect on liverfibrosis.2、In the development of nonalcoholic fatty liver thyroid hormone T3,T4values reducedgradually, TSH levels was increased.3、Exogenous thyroxine early intervention can effectively reverse the liver steatosis of ratswith nonalcoholic fatty liver, and there no obvious heart and liver side effects wereobserved.4、Advanced administration of exogenous thyroxine had no obvious effect on nonalcoholicfatty liver in rats with liver fibrosis.
