Objective:IL-33-induced AR mouse model and C-maf interference virus were used to explore the regulatory relationship among miR-155,C-maf and ILC2.Methods:1.Methods of establishing mouse AR modelFemale Balb/c mice 6 to 8 weeks old were selected and divided into an experimental group and a control group.The experimental group: the recombinant mouse IL-33(rm IL-33)0.5ug/20ul/mouse was intranasally administered on days 1,2,3.In the control group,the same volume of sterile PBS was administered.The mice were sacrificed on the 4th day,and the nasal mucosa tissues were collected to observe the histopathology through hematoxylin and eosin(HE)staining.2.The combined application of IL-33 and miR-155 agomir and antagomirMi R-155 agomir and miR-155 antagomir were intranasally treated 3 hours before the treatment of rm IL-33.The control mice received PBS by using same method.3.The use of lentivirusOn the first day,what the volume and concentration of LMRi82714 was intranasally administered were 20 ul and 9×108 TU/ml,and the next day,follow-up modeling was performed according to methods 1 and 2.4.Quantitative reverse transcriptase polymerase chain reaction(q RT-PCR)was used to detect the m RNA level of C-maf in the nasal mucosa of each group of mice.Enzyme-linked immunosorbent assay(ELISA)was measured the levels of IL-5 and IL-13 in the mucosa.Flow cytometry was used to detect the ratio of ILC2 in the nasal mucosa of mice.Results:1.The proportion of ILC2 in the nasal mucosa of mice in the AR group was higher than that in the WT group(P < 0.0001).Compared with the AR group,the proportion of ILC2 in the nasal mucosa of mice in the miR-155 agomir group was higher(P < 0.002).In contrast,the miR-155 antagomir group showed a downward trend(P < 0.0001).2.The levels of IL-5(P < 0.0001)and IL-13(P < 0.0001)in the nasal mucosa of mice in the AR group were higher than WT group.In the agomir group,IL-5(P < 0.0001)and IL-13(P < 0.0001)was higher than AR group,and the levels of IL-5(P < 0.0001)and IL-13(P < 0.0001)in the antagomir group were lower than AR group.3.LMRi82714,a C-maf interference lentivirus,can reduce the m RNA level of C-maf in nasal mucosa of mice(P < 0.002).In AR + LMRi82714 group,IL-5(P <0.0001)and IL-13(P < 0.05)in nasal mucosa of mice was higher than AR group.4.Compared with the antagomir group,the levels of IL-5(P < 0.0002)and IL-13(P < 0.05)in the nasal mucosa of the antagomir + LMRi82714 group were higher.Conclusion:1.Mi R-155 can up-regulate the amount of ILC2 in nasal mucosa of mice induced by IL-33.2.Mi R-155 can regulate the secretion of IL-5 and IL-13 in nasal mucosa of mice induced by IL-33 through C-maf.
