Objectives:Investigate the expression of hTERT,P16 and Ki67 in different grades of cervical intraepithelial neoplasia and cervical squamous cell carcinoma,explore the significa-nce of hTERT,P16 and Ki67 for triage of cervical intraepithelial neoplasia.Methods and Materials:1.Selected 48 patients who had been diagnosed with cervical lesions in the Second Hospital of Tianjin Medical University from March 2016 to April 2017,18-64years old age.All patients have been infected by high risk human papillomaviru?HR-HPV?and were diagnosed by biopsy or pathology postoperative.There were 12cases of chronic inflammation of cervical mucosa,28 cases of cervical intraepithelial neoplasia?CIN I 13,CIN II 9,CIN III 6?,and 8 cases of squamous cell carcinoma?SCC?.Surgery patients were not treated with chemotherapy before operation.2.Immunohistochemical staining of hTERT,P16 and Ki67 was performed in the most severe paraffin block.The positive expression rates of hTERT,P16 and Ki67were calculated respectively.The inflammatory group was used as a negative control,and the cervical cancer group was the positive control group.Result:1.The positive expression rate of hTERT,P16 and Ki67 was 0%?0/12?,0%?0/12?,9.09%?1/12?in inflammation group;15.38%?2/13?,23.08%?3/13?,30.77%?4/13?in CIN I;88.89%?8/9?,100%?9/9?,100%?9/9?in CIN II;100%?6/6?in CIN III;100%?8/8?in SCC.There was no significant difference in the expression of hTERT,P16 and Ki67 between the inflammation and the CIN I.The expression in CIN II,CIN III and SCC was significantly higher than the inflammation and CIN I groups,and the difference was statistically significant.There was no significant difference in CIN III and SCC.2.The correlation analysis of hTERT,P16 and Ki67 expression and cervical lesions shows that the expression of hTERT,P16 and Ki67 enhance with the increase of grade in cervical lesions,correlation coefficients were 0.879,0.905,0.870,P<0.005,showed that hTERT,P16 and Ki67 expression were positively correlated with cervical disease levels.3.Correlation analysis showed that P16 and Ki67,P16 and hTERT were positively correlated with the expression of cervical lesions in different grades(X2P16,Ki67=75.31,P<0.005,V=0.723,X2P16,hTERT=58.33,P<0.005,V=0.636).Conclusion:1.The expression of hTERT,P16 and Ki67 increased with the progression of cervical lesions and is positively correlated with the grade of cervical lesions.In clinical practice,the expression of hTERT,P16 and Ki67 should be detected in the tissues to determine the severity of cervical lesions in further,and identify the high grade CIN lesions,in order to triage the cervical precancerous lesions better,provide the guidance for clinical treatment.2.HTERT expression is closely related to cell proliferation and tumor occurrence.HTERT maybe predict CIN malignant potential combined with P16.It may become an auxiliary index in clinical for monitoring CIN's malignant potential and diagnosis of SCC and CIN.
