Objective:By studying the expressions and changes of TGF-?1 and OPN in gingival crevicular fluid(GCF)when the canines are moved distantly under four different orthodontic forces, the effects of different force values on their expression were discussed and whether TGF-?1 and OPN are involved in the reconstruction of periodontal tissue was learned, which is intended to summarize the optimal force for moving the canine in clinical practice and provide a theoretical basis for clinical application.Method:24 patients in jiangxi provincical people,s hosipital of Angle Class II Division 1with osteogenic class I and high-angle cases were selected and randomly divided into four groups(n=6).The first premolars on the left/right side of maxilla were extracted and four different forces were applied to the canines: 50 g in group A,100 g in group B,150 g in group C,250 g in group D.The other side was used as a control group without force. The GCF of the canine mesial site was collected before the force was applied and 1 d,7 d,21 d,28 d after the force was applied. The ELISA reagent method was used to detect OPN and TGF-?1 concentrations at different time points in the GCF and the canine movement distances were measured at each time point. The mean values of the TGF-?1 and OPN concentrations(x ± s)in the GCF at each time point were measured. The variances of TGF-?1 and OPN in GCF at different force values, time points, and locations was compared by analysis of variances of three-factor repeated measurement.P?0.05 was considered statistically significant.Results:1.The tooth movement distances at different time points and different force values had interactions with statistically significant differences(P <0.05);The tooth movement distances of each group under different force values were statistically significant(P<0.05);The tooth movement distance at different times under the same force value was also statistically significant(P <0.05).2.Among the three factors of force value, position, and time in the OPN measurement in GCF,except for position and force value(P>0.05),the others had interactions with statistically differences(P<0.05).3.Repeated measurement of TGF-?1 levels in GCF in the analysis of three-factors ANOVA showed that there were interactions among the three factors of time, force value, and position, and there was a statistically significant difference(P<0.05).Conclusions:1.In a healthy state,TGF-?1 and OPN are present in the gingival crevicular fluid and are closely related to the normal metabolism of periodontal tissues.2.Under different orthodontic forces, the expression of TGF-?1 and OPN changes have significant differences over time. When the force value is below 100 g,the changes of TGF-?1 and OPN levels are small, and the time required to recover to the baseline level is shorter. When the force value exceeds 100 g,especially under250 g force value, the levels of TGF-?1 and OPN were changed sharply, and the recovery time takes 4 weeks or more.
