Objective:By detecting T lymphocyte subsets(CD3~+T cells,CD4~+T cells,CD8~+T cells,Treg cells)and Cerebrospinal fluid cytokines(IL-2,IL-8,IL-10)in patients with viral encephalitis,We investigate the role of T lymphocyte subsets and Cerebrospinal fluid cytokines in the pathogenesis of viral encephalitis.Methods:A total of 25 patients diagnosed with viral encephalitis were selected as the viral encephalitis group in the Department of Neurology,Jiangxi Provincial People's Hospital from March 2019 to December 2019.In the same period,A total of 26healthy people who underwent physical examination in our hospital were selected as healthy control group.Among them,peripheral blood of patients with viral encephalitis were collected within 24 hours after their admission,peripheral blood were collected from healthy people from healthy controls on the same day.CD3~+T,CD4~+T,CD8~+T,Treg cells,CD4~+T/CD8~+T levels in peripheral blood were detected with flow cytometry.A total of 21 patients diagnosed with viral encephalitis were selected as the viral encephalitis group in the Department of Neurology,Jiangxi Provincial People's Hospital from September 2019 to January 2020.In the same period,A total of 23 patients diagnosed with non viral encephalitis in our hospital were selected as non viral encephalitis group.Lumbar puncture was performed within24 hours of admission in both groups.IL-2,IL-8 and IL-10 levels in cerebrospinal fluid were detected with multiplex flow cytometric immuno-bead array assay technology,and the changes of cerebrospinal fluid IL-2,IL-8 and IL-10 levels before and after viral encephalitis treatment were compared.SPSS 20.0 software was used to statistically analyze the laboratory test results of the two groups.Measurement data followed the normal distribution were analyzed by t test,otherwise were analyzed by non-parametric test.Counting data were analyzed by?2 test.Linear correlation analysis was used to analyze the correlation between cytokines in cerebrospinal fluid.Wilcoxon test was used to analyze the changes of cytokines in cerebrospinal fluid before and after treatment.If the P value was less than or equal to 0.05,the difference was considered statistically significant.Result:1.CD4~+T,CD8~+T,and Treg cells in peripheral blood of the viral encephalitis group were significantly higher than that of the healthy control group.2.The levels of cerebrospinal fluid IL-2,IL-8 and IL-10 in the viral encephalitis group were significantly higher than those in the non-viral encephalitis group.3.Cerebrospinal fluid IL-2 and IL-8,IL-2 and IL-10,IL-8 and IL-10 were positively correlated in the viral encephalitis group.4.The levels of IL-2 and IL-8 in the cerebrospinal fluid of the group after the viral encephalitis treatment were significantly lower than those in the group before the viral encephalitis treatment.There was no significant difference in the levels of IL-10 in the cerebrospinal fluid of the group after the viral encephalitis treatment compared with the group before the viral encephalitis treatment.Conclusion:The expression of CD4~+T,CD8~+T,Treg cells and cerebrospinal fluid IL-2,IL-8,IL-10 in the acute phase of viral encephalitis increased,which may be involved in the development of viral encephalitis and mediate immune response.As inflammation gradually subsided,cerebrospinal fluid IL-2 and IL-8 decreased.Therefore,in viral encephalitis,the increased expression of CD4~+T,CD8~+T,Treg cells and cerebrospinal fluid IL-2,IL-8,IL-10 is helpful for the diagnosis of viral encephalitis,and IL-2 and IL-8 can be used as evaluation indicators for the efficacy of viral encephalitis.
