Objective: The object of this article is to investigate the relationship between Dentonin and bone mass in periodontitis,and the effect of Dentonin on osteoblast proliferation.Methods: Eighteen SD rats were randomly divided into three groups: normal group,model group and dexamethasone group.In the model group and dexamethasone group,10 ?l of normal saline containing 1 mg/ml E-LPS was injected into the buccal and palatal gingival sulcus beside the maxillary first molar of the rats on the right side to establish experimental rat model of periodontitis.Dexamethasone group were treated with 40 ?mol/L dexamethasone once every 48 hours for 3 times.The alveolar bone was surgically removed,and the expression of Dentonin was detected by qPCR.Rat mandibular osteoblasts were extracted and different concentrations(0,0.04,0.4,4,40 ?mol/L)of Dentonin were applied to osteoblasts.On the 7th day of culture,the proliferation of osteoblasts was detected by Cell Counting Kit-8(CCK-8)method,and the activity of alkaline phosphatase(ALP)was detected.Expressions of P-I?B? and I?B? were detected by Western blotting.Results: qPCR results showed that the expression of Dentonin in the normal group(0.515±0.404)was significantly lower than that in the normal group(2.744±0.955)(P<0.05),which was statistically significant.Compared with the model group(0.515±0.404),the dexamethasone group promoted Dentonin expression in periodontal tissue(1.480±0.447)(P=0.086).After 7 days of treatment of Dentonin,CCK-8 results showed that the OD values of 0,0.04,0.4and 4 ?mol/L groups were(0.287±0.018),(0.296±0.021),(0.373±0.043)(P < 0.05)and(0.583±0.046)(P<0.01).The ALP activities in the 0,0.04,0.4,4,and 40 ?mol/L groups were(133.207±53.265),(150.884±91.570),(155.934±175.808),(277.778±34.409)(P<0.01),(278.409±98.266)(P<0.05),and higher concentration of Dentonin led to higher ALP activity.Western Blot showed that the expression of P-I?B? protein was significantly reduced in the 4 ?mol/L group.Conclusions: Dentonin is involved in the regulation of bone mass in periodontitis,and can promote the proliferation and differentiation of osteoblasts by inhibiting the NF-?B signaling pathway.
