Objective:To evaluate the changes of Wnt3a,Dkk1in alveolar bone on experimental perodontitis, and further explore the effect and mechanism of wnt signaling pathway in experimental periodontitis in rats.Methods:sixty-four (6to8-week-old) male rats were randomly divided into two groups, control group (n=32), model group (n=32), the periodontitis group includes32rats by co-ligation with silk thread in the mouth and inoculation of bacteria. After rats models were established, animals were sacrificed after2wk,4wk and6wk. Obtain samples from rat maxilla,4%paraformaldehyde, HE staining, TRAP staining and Wnt3a,Dkk1in immunohistochemistry. Observed indexes include:(1) Clinical index: probing depth (PD);(2) animals were killed after the blood through the heart, centrifugation, ELISA to detect serum in Wnt3a,Dkk1protein level changes;(3) histologic index:HE stained histology, TRAP staining of inflammatory alveolar crest1/3broken bones cell counts, immunohistochemical staining of inflammatory alveolar ridge top1/3of bone cells, to observe the change of optical density of the Wnt3a,Dkk1positive expression; Data was analyzed by paired t-test and single factor analysis of variance statistically.Results:Periodontal tissue inflammation gradually severe in periodontitis model group With the increase of the modeling time. periodontal probe index (PD) increases gradually in Periodontitis model group, osteoclast count also increased with the increase of time at the unit length oflnflammatory alveolar ridge; Compared with the normal group, the optical density value of Wnt3a positive expression decreases in inflammatory alveolar ridge top1/3of bone cells while the optical density value of Dkk1increases. Wnt3a levels gradually decreases in peripheral blood serum while Dkkl levels increases.Conclusions:with the inflammation gradually severed in periodontitis model group, we found the Wnt signaling factor Wnt3a gradually reduced while the Wnt signaling factor Dkkl increases. Wnt3a signaling pathway may play an important role in experimental periodontitis in rats.
