Objective:To detect the expression of TRPA1 and TGF-β1 in epulis and normal gingival tissue. To analyze the relationship between TRPA1 and TGF-β1 and the clinicopathological parameters of epulis and the correlation between TRPA1 and TGF-β1 expression in epulis.Methods:201 cases of epulis archived paraffin specimens(Gingival tumor group)were surgically removed from June 2008 to June 2018 in the First Affiliated Hospital of Shihezi University,Shihezi City.A total of 48 cases of gingival tissue after impacted extraction of mandibular third molars in a department of dentistry of an affiliated hospital(normal gingival group). The expression of TRPA1 and TGF-β1 in the two groups was detected by immunohistochemistry, and the correlation between the two proteins and the development of epulis was statistically analyzed by software SPSS 23.0.Results:The expressions of TRPA1 and TGF-β1 in the squamous epithelium were in the cytoplasm of the epithelial basal layer, spinous layer, granular layer and lamina propria in order from deep to shallow, and mainly expressed in vascular endothelial cells in the stroma. The cytoplasm and cell membrane of fibroblasts were light yellow or brown, and a small amount was expressed in the nucleus. The positive rates of TRPA1 in normal gingival tissue and gingival tumor tissue were 35.42%(17/48)and 56.72%(114/201),the difference was statistically significant((34)(17)=11.125,P<0.05).The positive expression rates of TGF-β1 in normal gingival group and gingival tumor group were 25.00%(12/48)and 90.05%(181/201),and the difference was statistically significant((34)(17)=96.519,P<0.05).The expression of TRPA1 was significantly related to the age of patients with epulis((34)(17)=12.632,P<0.05),and the older the age,the lower the expression of TRPA1.There was no significant difference in the expression of TGF-β1 in the age of epulis patients(P>0.05).The positive rate of TRPA1 and TGF-β1 in non-drug-induced gingival overgrowth was significantly higher than that of drug-induced gingival overgrowth, and the difference was statistically significant((34)(17)=48.921 and(34)(17)=12.197,P<0.001).There was no significant difference in the expression of TRPA1 and TGF-β1 in terms of gender and ethnicity(P>0.05).The expression of TGF-β1 was correlated with pathological classification((34)(17)=18.103,P<0.05).The highest positive rate of angiogenic epulis was 97.37%(37/38),followed by fibrous epulis95.37%(103/108),and granulation The positive rate of tumourous epulis was lower 74.55%(41/55).There was a significant positive correlation between TRPA1 and TGF-β1 expression in epulis tissues(r=0.507,P<0.001).Conclusion:(1)There is a significant positive correlation between TRPA1 and TGF-β1expression in epulis, and there is a synergistic effect between the two;(2)TRPA1 has an inhibitory effect on drug-induced gingival overgrowth in epulis. |
