Objective To investigate the relationship between single nucleotide polymorphisms(SNPs)at rs150846,rs8065080,rs161385 and rs222747 of TRPV1 gene and susceptibility to asthmatic diseases in Han,Uygur and Kazak children in Xinjiang.Methods In a case-control study,248children with asthmatic diseases were selected as the case group,including 94 Han nationality,49 Uygur nationality and 105 Kazak nationality,and the other 191 children were selected as the control group,including 60 Han nationality,53 Uygur nationality and 78 Kazak nationality.The venous blood 2 ml was extracted and DNA,was extracted to detect the genotypes of rs150846,rs8065080,rs161385 and rs222747 of TRPV1 gene by Massarray SNP typing method.Using SPSS 20.0 to sort out the data,the measurement data that do not conform to the normal distribution are expressed by Median(Quartile),and the differences between groups are expressed by rank sum test;?~2 test and conditional Logistic regression analysis are used to compare the genotype frequency and allele frequency distribution of rs150846,rs8065080,rs161385 and rs222747 of TRPV1 gene,and calculate the relative risk(OR)and 95%confidence interval(95%CI).The distribution of genotypes in the case group and the control group was tested by Hardy-Weinbery equilibrium law.The difference was statistically significant(P<0.05).Results(1)Comparison of general data:there was no significant difference in ethnic distribution,sex ratio and age between the case group and the control group(P>0.05).(2)The rs150846,rs8065080,rs161385 and rs222747 of TRPV1 gene in the case group and the control group were in accordance with the law of Hardy-Weinbery equilibrium.(3)The rs150846 genotype and allele frequency distribution of TRPV1 in the case group and the control group were analyzed.The genotypes were AA type(65 cases),GG type(172cases)and GA type(202cases).The frequencies were 14.8%,39.2%,46.0%,the A allele frequency was 37.8%,and the G allele frequency was 62.2%.There was no significant difference in the distribution of genotypes and alleles at rs150846 locus between the case group and the control group(P>0.05).The rs8065080 genotypes were CC type(133cases),TT type(84cases)and TC type(222cases).The frequencies are 30.0%,19.1%,50.6%,The T allele frequency was 44.4%,and the C allele frequency was55.6%.There was no significant difference in the distribution of genotypes and alleles at rs8065080 locus between the case group and the control group(P>0.05).The rs161385 genotypes were CC type(121cases),GG type(91cases)and GC type(227cases).The frequencies were 27.6%,20.7%,51.7%,The C allele frequency was 53.4%,and the G allele frequency was 46.6%.There was no significant difference in the distribution of genotypes and alleles at rs161385 locus between the case group and the control group(P>0.05).The rs222747 genotypes were CC type(101cases),GC type(211cases)and GG type(127cases).The frequencies were 23.0%,48.1%,28.9%.The C allele frequency was 56.5%,and the G allele frequency was 43.5%.There was no significant difference in the distribution of genotypes and alleles at rs222747locus between the case group and the control group(P>0.05).(4)There was no significant difference in the frequencies of genotypes and alleles at rs150846,rs8065080,rs161385 and rs222747 between the Han case group and the control group(P>0.05).(5)There was no significant difference in the distribution frequency of genotypes and alleles at rs150846,rs8065080,rs161385 and rs222747 between the Uygur case group and the control group(P>0.05).(6)Among the Kazakh population,The patterns of genotype and allele frequencies of rs150846 at the TRPV1 locus had statistical difference between the groups(P<0.05).The allele A of rs150846 had a 1.64-fold(95%CI:1.058-2.543)increased risk of developing asthmatic diseases compared with those who had allele G.(7)There was no significant difference in the distribution of genotypes and alleles at rs8065080,rs161385 and rs222747 between the Kazakh case group and the control group(P>0.05).(8)In the case group,there was no significant difference in the distribution of genotypes and allele frequencies of rs150846 and rs8065080 of TRPV1 gene among Han,Uygur and Kazakh populations,but there were significant differences in genotypes and allele frequencies of rs161385 and rs222747 of TRPV1 gene among Han,Uygur and Kazak populations.(9)There were significant differences in genotype and allele frequency distribution of rs161385 and rs222747 between Han nationality and Uygur nationality,and between Han nationality and Kazak nationality in rs161385 and rs222747(P<0.05).However,there was no significant difference in the distribution of genotypes and alleles at rs161385 and rs222747 between Uygur and Kazak(P>0.05).Conclusions(1)The rs150846 gene polymorphism of TRPV1 gene could be associated with susceptibility to asthmatic disease in children of Kazakh in Xinjiang.(2)There may be ethnic differences in the rs161385 polymorphisms of TRPV1 gene among Han,Uygur and Kazak children with asthmatic diseases in Xinjiang.(3)There may be ethnic differences in the rs222747 polymorphisms of TRPV1 gene among Han,Uygur and Kazak children with asthmatic diseases in Xinjiang.
