Objective:To simulate the clinical pathological process of human intracerebral hemorrhage(ICH)by establishing rat models with ICH.Recombinant human erythropoietin(rhEPO)was used to intervene the model rats,so as to explore whether rhEPO can improve the cognitive dysfunction of rats with intracerebral hemorrhage and its effect on the expression of apoptosis related factors Bcl-2 and Bax in hippocampusMethods:1.Fifty-four healthy male Sprague-Dawley rats were randomly divided into sham operation group(Sham group,18 rats),cerebral hemorrhage group(ICH group,18 rats),cerebral hemorrhage+recombinant human erythropoietin group(rhEPO group,18 rats),and each group was further divided into three subgroups according to the intervention time(the 3rd day,7th day and 14th day),with 6 rats in each subgroup2.Three-dimensional stereotaxic technique and intracerebral hemorrhage model induced by collagenase injection were used to prepare experimental rat model with intracerebral hemorrhage in rhEPO group and ICH group.In rhEPO group,rhEPO(rhEPO 5000U/kg,once a day)was injected subcutaneously into the abdomen,while in ICH group,normal saline(once a day)was injected into the abdomen at the same time,at the same site and at the same dose.There was no treatment in the sham group3.The Morris water maze(MWM)test was carried out on the 3rd,7th and 14th day after intervention in each group,including place navigation test and spatial probe test to test the learning and memory function of rats with intracerebral hemorrhage.After MWM test,the hippocampus of rats was collected,immunohistochemistry(IHC)was used to detect the expression levels of apoptosis-related factors Bcl-2 and Bax in rat hippocampal tissues of different groups;the apoptosis level of rat hippocampal tissue was detected by TUNEL methodResults:1.MWM test results:?Compared with sham group,rhEPO group had longer escape latency on the 3rd,7th and 14th day(P<0.05);compared with ICH group,rhEPO group had shorter escape latency on the 3rd,7th and 14th day(P<0.05);?compared with sham group,rhEPO group had fewer times of passing through the platform on the 3rd,7th and 14th day(P<0.05);compared with ICH group,rhEPO group had more times of passing through the platform on the 3rd,7th and 14th day(P<0.05)2.Detection results of IHC:? the expression level of Bcl-2 in rhEPO group was significantly lower than that in sham group on the 3rd,7th and 14th day(P<0.05);the expression level of Bcl-2 in rhEPO group was significantly higher than that in ICH group on the 3rd,7th and 14th day(P<0.05),and the expression level of Bcl-2 in rhEPO group was the lowest on the 3rd,increased on the 7th day and the highest on the 14th day(P<0.05);? the expression level of Bax in rhEPO group was significantly higher than that in sham group on the 3rd,7th and 14th day(P<0.05);the expression level of Bax in rhEPO group was significantly lower than that in ICH group on the 3rd,7th and 14th day(P<0.05),and the expression level of Bax in rhEPO group was the highest on the 3rd day,decreased on the 7th day and the lowest on the 14th day P<0.05)3.Detection results of TUNEL method:the number of TUNEL positive cells in rhEPO group was higher than that in sham group on the 3rd,7th and 14th day(P<0.05).The number of TUNEL positive cells in rhEPO group was lower than that in ICH group on the 3rd,7th and 14th day(P<0.05),and the number of TUNEL positive cells in rhEPO group was the most on the 3rd day,decreased on the 7th day and the least on the 14th day(P<0.05)Conclusion:1.rhEPO can improve the cognitive function of rats with experimental intracerebral hemorrhage;2.rhEPO can promote the expression of anti-apoptosis factor Bcl-2 in the hippocampus of rats with ICH,and inhibit the expression of pro-apoptosis factor Bax and the apoptosis of hippocampus.
