Objective: The experiment was to study the relation of cell apoptosis and the expression of c-fos at perihematomal brain tissue at different time after intracerebral hemorrhage, to explore the mechanisms of secondary injury after ICH, and to provide the theoretical basis for the treatment of intracerebral hemorrhage. Methods: 1. Forty-two adult rabbits weighed 2.5~3.0kg without limitation of sex were randomly divided into normal group and experimental group. Group A was normal group. Experimental group was divided into six subgroups with different time randomly(6 rabbits per group). Models of intracerebral hemorrhage were made by injecting autoblood into brain tissues of rabbits. 2. The level of c-fos mRNA were determined by means of RT-PCR method, the level of c-fos protein were determined by means of immumnohistochemical SP method in the perihematomal brain tissues, and the level of apoptosis were determined by means of TUNEL method. 3. Statistical analysis was done with SPSS13.0 statistical package. Results: 1. The normal group have no apoptosis and the expression of c-fos mRNA and c-fos protein. The experimental group have apoptosis cell and the expression of c-fos. 2. The peak of c-fos mRNA expression appeared at 0.5h after ICH, degraded gradually until disappeared at 24h. The c-fos protein appeared at 0.5h, rised to peak at 2h, and degraded gradually. Adversely, the apoptosis cell increased gradually, and rised to peak at 48h. Conclutions: 1. Making intracerebral hemorrhage model with injecting auto-blood is satisfied and reliable. 2. Apoptosis, c-fos mRNA and c-fos ptotein of experimental group and the normal group have significant difference. 3. c-fos gene is related to apoptosis in perihemotomal brain tissues, and cause cell apoptosis after intracerebral hemorrhage..
