Objective:Liquid chromatography mass spectrometry(LC-MS)was used to detect the urine of patients with early gastric cancer,patients with advanced gastric cancer,and normal people.The differences in the metabolites in the urine of the three groups were analyzed,and then the early gastric cancer was predicted and evaluated Potential biological markers of gastric cancer progression.Method:1.The experimental groups were divided into early gastric cancer group and advanced gastric cancer group according to preoperative examination and postoperative pathology.In addition,a group of healthy volunteers was selected as a control based on the medical report.2.Sample processing: After removing the sample at room temperature,thawing,adding acetonitrile,high-speed centrifugation(14000rpm,5min),taking the supernatant for detection and analysis by HPLC-MS.3.Chromatographic analysis and data processing: Three groups of urine samples were detected by liquid chromatography-mass spectrometry,and metabolite spectra of early gastric cancer group,advanced gastric cancer group,and normal people were obtained.Further use Matlab software to normalize the original data and do principal component analysis,and then use statistical methods such as analysis of variance and partial least squares discriminant analysis to find important relationships between early gastric cancer patients,advanced gastric cancer patients,and normal people.Differential metabolites were further screened by cluster analysis and ROC curve drawing,and differential metabolites with strong discrimination ability were identified,and the structure of differential metabolites was identified through the human metabolome database.Result:through the total ion flow chart of the three groups of urine samples,it can be intuitively observed that there are differences in metabolites between the three groups of urine samples.The results of the principal component analysis chart show that the three groups of urine samples are well dispersed and can be distinguished well.It is further confirmed that there are differences in the metabolites of the three groups of urine samples.together.A total of 324 differential metabolites were screened out by analysis of variance(P <0.05).Eleven important metabolites were further screened using partial least squares discriminant analysis.Data were imported into the human metabolome database to find 6 differential metabolites.Finally,through cluster analysis and drawing ROC curve,five different metabolites with strong discrimination ability were further screened.Conclusion:1.Metabolites in urine of early gastric cancer group and healthy group are different,7-Methylguanine,Vinylacetylglycine,4-Vinylphenolsulfate may be potential biomarkers of early gastric cancer;2.The metabolites in urine of gastric cancer group and healthy group are different.The differential metabolites are Butyric acid,4-Vinylphenolsulfate and 7-Methylguanine respectively;3.7-Methylguanine showed a gradual increase in the healthy group,early group and advanced gastric cancer group;Butyric acid showed a downward trend in the healthy group and gastric cancer patients,and the urine content in early gastric cancer was lower than that in advanced gastric cancer.The content of 7-Methylguanine and Butyric acid in human urine may be related to the occurrence and development of tumors.
