Objective: To compare,using HPLC-MS,the differences across the early gastric cancer group,chronic superficial gastritis and health people group in the serum and urine low-molecular-weight metabolites,in search of the metabolites that may contribute to the diagnosis of early gastric cancer.Methods: Serum and urine specimens were collected from 16 patients with early gastric cancer,20 patients with chronic superficial gastritis and 20 health people in Baise Municipality of Guangxi Zhuang Autonomous Region with high prevalence of gastric cancer.Samples collected from each group were tested using HPLC-MS;then principal component analysis(PCA),partial least squares discriminant analysis(PLS-DA)and t-test were performed to determine the differential metabolites for the early gastric cancer group and the rest two groups;finally the differential metabolites were identified by searching the Human Metabolome Database(HMDB).Results: In comparison with health people,9(Dihydrogen cholesterol,Cholesterol,Tyrosine,Lactic acid,Tyramine,3-hydroxypropionic,Alanine,LPC(16:0),Pyruvate)and 8(Hexadecanoic acid,Lactic acid,Pyruvaldehyde,Cholesterol,Valine,Glycerol,Inositol,Aspartic acid)metabolic markers that may contribute significantly to disease differentiation were identified in the serum and urine,respectively,of patients with early gastric cancer.In comparison with patients chronic superficial gastritis,9(L-Valine,Cholesterol,L-carnitine,Ornithine,Docosahexaenoic acid,Pyruvaldehyde,Lysophosphatidylcholine(16:0)and 7(Succinic acid,Propionic acid,Serine,L-Isoleucine,cholesterol,2-hydroxybutyric,Methionine)metabolic markers that may contribute significantly to disease differentiationwere identified in the serum and urine,respectively,of patients with early gastric cancer.Changes in these metabolites are indicative of the metabolic disorders of carbohydrates,lipids and amino acids in patients with early gastric cancer.Conclusion: Metabolomics analysis based on HPLC-MS is valid for differentiating early gastric cancer from chronic superficial gastritis and health people,indicating its potential diagnostic value for early gastric cancer.
