Mongolian medicine Sulongga-4 Granules?SG?is an innovative drug developed on the based of Sulongga-4 decoction.Compared with decoction,the granule is small in size,easy to carry,store and transport,non-perishable and good in taste,with high patient compliance,which makes it more popular than decoction.It is composed of four herbs of Forsythia suspensa,Polygonum bistorta L,Akebia quinata and Ophiopogon japonicus.It is cool in nature and has the effect of clearing viscera heat.It is mainly used for intestinal heat,dysentery,abdominal pain and diarrhea.Rarely make the news to its pharmacological research,the purpose of this study is to explore the anti-diarrhea of SG,astringent,anti-inflammatory,analgesic and antipyretic effects,and the method based on network pharmacology comprehensive projections for the mechanism of action of SG anti diarrhea,according to the forecast results in animal experiments to further explore the mechanism of action of SG anti diarrhea,on the whole evaluation of SG role at the same time,the mechanism of action of the anti diarrhea also provide a theoretical basisExperimental results:1.Results of pharmacodynamic experiments?1?Antidiarrheal test results show that SG has a significant antidiarrheal effect on diarrhea caused by irritating laxatives senna and castor oil and can significantly delay the occurrence of diarrhea and reduce the symptoms of diarrhea in mice;The antidiarrheal effect of diarrhea is the least obvious,but it can significantly delay the onset of diarrhea.?2?The results of mice small intestinal propulsion experiments showed that SG significantly inhibited intestinal propulsion in normal mice and intestinal propulsion in diarrhea model mice caused by castor oil,and its effect was similar to that of the positive drug loperamide.?3?Anti-inflammatory test results show that SG can significantly reduce the swelling rate of ears caused by xylene and increase the swelling inhibition rate.?4?The results of analgesic experiments showed that each dose group of SG could significantly reduce the number of writhing reactions in mice caused by glacial acetic acid and increase the rate of writhing suppression.?5?The results of antipyretic experiments showed that the body temperature of rats in the dry yeast group at a concentration of 1.5g·kg-1 increased fastest,and reached a maximum at 7 hours;SG had a significant inhibitory effect on the body temperature of fevered rats.2.Results of network pharmacology experiments?1?SG has a total of 34 active ingredients and 110 targets,diarrhea has a total of 3,500 known therapeutic targets,SG has 74 anti-diarrhea targets,of which 28 are core targets.?2?Analysis of the GO biological process yielded 87 entries,and analysis of the KEGG pathway yielded 117 pathways,which mainly involved PI3K-Akt signaling pathway,TNF signaling pathway,p53 signaling pathway,and apoptosis signaling pathway.3.Experimental results of mechanism exploration?1?SG can significantly increase the intestinal villi length and increase V/C value.?2?The results of inflammation-related factors showed that SG can down-regulate the expression of RELA,IL-6,TNF-?,and ICAM-1 in small intestinal epithelial cells of diarrhea-induced diarrhea mice.?3?The results of apoptosis-related factors showed that SG can down-regulate the expression of CASP-8 and CASP-3 in small intestinal epithelial cells of diarrhea-induced diarrhea mice.Conclusion:The efficacy experiments in this study show that SG has certain antidiarrheal,astringent,anti-Inflammation,analgesic and antipyretic effect.In the network pharmacology-based experiments to predict the possible mechanism of SG anti-diarrhea,it shows that SG anti-diarrhea presents multiple targets and multiple pathways,which comprehensively suggests the possibility of SG anti-diarrhea.It is predicted that SG may block the inflammatory response by inhibiting the production of inflammatory factors and affect the apoptosis of the intestinal mucosa while repairing it,and finally play an anti-diarrheal effect.Mechanism investigation experiments show that SG reduces inflammation by regulating the inflammatory factors IL-6,TNF-?,RELA and ICAM-1,and regulating the apoptosis factors CASP-8 and CASP-3 to inhibit apoptosis in order to protect the villus,Damage to the crypts,thereby improving the symptoms of diarrhea.The findings provided experimental references for the clinical application of SG.The network pharmacological approach provided new ideas for the development of new medicines for modern ethnic medicine and its compounds.The related research findings also provided a scientific basis for explaining the treatment of diarrhea by SG.
