| Purpose:This study intends to explore the active components and targets of anti-atherosclerosis in Tongmai Jiangzhuo recipe based on the method of network pharmacology,and predicts its potential mechanism and material basis.In addition,it intends to provide evidence for the clinical efficacy of Tongmai Jiangzhuo recipe and provide the direction for the follow-up in-depth experm ental research.Material and method:The potential active components of 12 traditional Chinese medicines in Tongmai Jiangzhuo recipe were screened according to oral bioavailability≥30%and drug-like≥0.18 in TCMSP database.I the meanwhile,the single anti-atheroscl erotic chemical components in the 12 traditional Chinese medicines of Tongmai Jiangzhuo recipe were screened by literature mining of CNKI,VIP and Wanfang database The related targets of the potential active components were obtained through the TCMSP database,and the targets were transformed into corresponding gene names through the UinPort database.Moreover,the smile numbers of the chemical components not retrieved by TCMSP were obtained by the PubChem database,and the related target genes were predicted by the Swiss Target Prediction database.In addition,the target genes of atherosclerotic(AS)were obtained from the Drugbank databases of GeneCards,OMIM,and TTD.The key target genes were obtained by mapping the target genes of Tongmai Jiangzhuo recipe and AS disease target genes.Besides,the traditional Chinese medicine-component-key target network was constructed by Cytoscape3.6.1 software,the PPI network of key target genes was drawn based on STRING,and the PPI network was analyzed by topology analysis and MCODE module cluster analysis based on Cytoscape3.6.1 software.The key target genes were analyzed by GO enrichm ent analysis and KEGG enrichment analysis through DAVID database,and the data were visualized through Yihaabo bininformatics mapping website.Results:1.302 potential active components of Tongmai Jiangzhuo recipe were obtained based on TCMSP database and literature mining,including 19 from Bupleurum,23 from Fructus Aurantii,37 from Scutellaria baicalensis,13 from Pinellia ternata,13 from Radix Paeoniae Alba,68 from Radix Salviae Miltiorrhizae,12 from Panax notoginseng,17 from Rhubarb,12 from Alisma orientalis,95 from Licorice,13 from Polygonum cuspidatum and 7 from Rosa laevigata Michx.2.753 target genes related to the anti-atherosclerosis potential active components of Tongmai Jiangzhuo recipe were obtained by TCMSP database,UinPort database and Swiss Target Prediction database.3.4375 target genes of atherosclerosis were obtained based on GeneCards,OMIM,Drugbank and TTD databases.4.532 key target genes of Tongmai Jiangzhuo recipe against AS were obtained after mapping the target gene data set of Tongmai Jiangzhuo recipe and the disease target gene dataset.5.15 possible key chemical components of Tongmai Jiangzhuo recipe against AS were obtained through the analysis of medicinal material-component-key target network,such as quercetin,ginsenoside Rg1,glycyrrhizic acid,Salvianolic acid B and so on.6.Three potential functional clusters of Tongmai Jiangzhuo recipe against AS were obtained by analyzing the key target gene PPI network.In addition,GO-BP analysis showed that the three clusters played anti-atherosclerosis effects in different ways,such as participating in negative regulation of amine transport,multiple regulation of cell cycle,regulation of protein kinase activity and so on.7.The results of GO enrichment analysis of key target genes:According to GO-CC results.the effective components of Tongmai Jiangzhuo recipe against AS may have biological functions in intercellular space,cytoplasm,plasma membrane,exocrine body,mito-chondrial outer membrane and so on.GO-MF results show that Tongmai Jiangzhuo recipe can play a role in anti-atherosclerosis by regulating protein het erodimerization activity.ATP binding and steroid receptor active molecular function.GO-BP results showed that Tongmai Jiangzhuo recipe may be involved in inflammaatory reaction,apoptosis signal pathway,cytoplasmic calcium concentration,redox process,glucose and lipid metabolism and so on to interfere with the progression of AS.8.The results of KEGG enrichment analysis of key target genes showed that Tongmai Jiangzhuo recipe may have anti-atherosclerosis effect by regulating PI3K-Akt signal pathway,FoxO1 target,MAPK signal pathway,NF-κB signal pathway,mTOR signal pathway,Jak-STAT s signal pathway and so on.Conclusion:1.Quercetin,ginsenoside Rg1,glycyrrhizic acid,Salvianolic acid B,notoginsenoside R1,glycyrrhetinic acid,emodin,24-acetyl alisol A,saikosaponin D,ginsenoside Rb1,hesperidin,glycyrrhizin,kaempferol,β-sitosterol and wogonin are the key active components in Tongmai Jiangzhuo recipe.2.The anti-atheroscl erosis mechanism of Tongmai Jiangzhuo recipe is realized through multi-targets and multi-pathways.It may involve inflammatory reaction,oxidative stress,cell proliferation,migration and apoptosis,glucose and lipid metabolism and other biological processes,as well as PI3K-Akt,FoxO1,MAPK,NF-κB,mTOR,Jak-STATs and other signal pathways. |
