| Tang Minzhi,the founder of Tonghua Deyi Hall,created the formula of Xueshuantongmai capsule according to the ancient formula of Yizongjinjian and combined with his own clinical practice.It has the functions of invigorating Qi,activating blood circulation,dredging collaterals,resolving phlegm,and removing blood stasis.However,the traditional decoction has many problems,such as cumbersome preparation process,incomplete extraction of effective ingredients,and carrying difficulty.In this paper,from the standpoint of standardizing drug production,improving drug quality,reducing patients.dosage and being convenient to carry.On this basis,the Xueshuantongmai capsule was prepared by modern technology.Xueshuantongmai capsule’s main ingredient is Astragali Radix,supplement ingredients are Pheretima,Eupolyphaga Steleophaga,Hirudo,Persicae Semen,Chuanxiong Rhizoma,Citri Exocarpium Rubrum,Angelicae Sinensis Radix and Spatholob Caulis.This prescription uses Astragali Radix to reinforce vital energy.Qi is vigorous,and blood circulation will be better,then the thrombus will be dissolved.Insect drugs can dissolve thrombus and remove blood stasis.Angelicae Sinensis Radix can promote blood circulation and nourish blood.Citri Exocarpium Rubrum to dissolve phlegm.Spatholob Caulis to dredge collaterals.The whole prescription has the effect of activating blood circulation,dissolving thrombus,relaxing channels,dredging collaterals,can be used to treat cardiovascular and cerebrovascular thrombus.Xueshuantongmai capsule to kes Astragali Radix as the monarch medicine,so Astragaloside A as the final evaluation index.In consideration of the solubility of the effective ingredients in the prescription,the ethanol reflux method was used to extract the medicinal materials in the experiment,and the optimal extraction process was determined through the orthogonal test-immerse the medicinal materials overnight with 8 times of the solvent amount of 70%ethanol.Heated and refluxed for 1 h.Extracted twice,and liquid filtration.Then use the rotary evaporator to evaporate the liquid.When the liquid is slightly viscous,put the liquid into the evaporating dish.The extractum was evaporated in water bath at 75℃ and dried at 55℃ under reduced pressure.Then study the preparation forming process,and determine the appropriate subsidiary materials,wetting agent and proportion of extractum powder and subsidiary materials(proportion of extractum powder and starch is 6:4,spraying 85%ethanol to wet,kneading into a group,mechanical pressing through 20 mesh sieve,drying at 40℃,then passing 40 mesh sieve,removing dust,filling and packaging).Through thin-layer identification and high performance liquid chromatography of each component of Xueshuantongmai capsule,and according to the requirements of capsule in the general principles of four part preparations of Chinese Pharmacopoeia(2015 Edition),carry out corresponding item inspection and establish quality standards.The results showed that the spots of the sample and the control medicine/standard sample were at the same position in TLC,but the negative control sample had no corresponding spots in the same position.The content of the effective components in the test was high.The difference of the loading amount,the water content detection,disinteg ration time limit under the capsule requirements were all in accordance with the provisions of the Chinese Pharmacopoeia.According to the standard of the 9001 capsule requirements in the general principles of four part preparations of Chinese Pharmacopoeia(2015 Edition),the influencing factors test was carried out.The results showed that the stability of Xueshuantongmai capsule was good.Xueshuantongmai capsule was used to oral administration to rats respectively.After 7 days of continuous administration,blood was collected from the abdominal aorta to get the serum containing medicine.The serum containing medicine was used to protect HUVEC and PC12 cells,and then H2O2 was used to oxidize HUVEC and PC12 cells to detect the protective effect of the serum containing medicine on HUVEC and PC12 cells.The results showed that the indexes of oxidative damage were significantly reduced,indicating that the serum containing medicine had the effect againist antioxidant damage.After 7 days of continuous oral administration,the rats were treated with FeCl3 to make thrombus model.The thrombus weight,inhibition rate of thrombus formation and other clotting factors were detected.After 7 days of continuous oral administration,the mices’ clotting time and bleeding time were detected.The results showed that Xueshuantongmai capsule could effectively improve the symptoms of thrombosis rats/mices. |
