Objective By observing the effects of acupuncture on the pathomorphology,apoptosis and cytokine expression of hippocampus in rats with epilepsy states,the specificity of acupoint effect in single and group points was explored.Method 120 SD rats were randomly divided into 12 groups,namely the healthy group(healthy blank,healthy meridian,healthy control,healthy Baihui,healthy Dazhui,healthy Zusanli)and the disease group(disease blank,disease meridian,disease Control,disease Baihui,disease Dazhui,disease Zusanli)10 rats in each group,rats in the disease group used the epilepsy rat model.After the successful modeling,each group was intervened for 10 days,once a day for 30 minutes each.The treatment period was 10 days,and samples were taken on the 11 th day.Take the left hippocampus of each group of rats and observe the pathological morphological changes of the neurons with HE staining;take the right hippocampus of each group of rats and observe the apoptosis and cells of the hippocampus by flow cytometry and liquid chip technology Factor expression.Result 1.Hippocampal tissue morphology results:(1)In the state of rat healthy function:Compared with the healthy blank group,the rate of positive cells in the hippocampus tissue of acupuncture groups of healthy Dazhui,Baihui,Zusanli single point group,healthy meridian point group and healthy control group Compared with the healthy meridian point group,the positive cell rate of each single point group of healthy Dazhui,Baihui and Zusanli decreased(P<0.05),and the positive cell rate of healthy control group increased(P<0.05).(2)In the state of rat disease function: Compared with the blank group,the acupuncture disease Dazhui,Baihui,Zusanli single point group,disease meridian point group and disease control group hippocampus tissue positive cell rate showed a decreasing trend(P<0.05);Compared with the disease meridian acupoint group,the positive cell rate in the single acupoint groups of the disease Dazhui,Baihui and Zusanli in the hippocampus DG area decreased(P<0.05).2.Apoptosis test results:(1)In the state of rat healthy function: Compared with the healthy blank group,the apoptotic rate of early and late neurons in the hippocampus of the acupuncture healthy meridian group and healthy control group increased(P<0.05);Compared with the healthy meridian group,the early apoptosis rate in the healthy Zusanli group increased(P<0.05),and the late apoptosis rate in the healthy Dazhui group increased(P<0.05).(2)In the disease state of rats: Compared with the blank group of the disease,there was no statistical significance in the early cell apoptosis rate of the acupuncture disease Dazhui,Baihui,Zusanli single point group,disease meridian point group and disease control group(P>0.05);compared with the disease meridian group,the late-stage apoptosis rate of the disease Dazhui group and the disease Baihui group increased(P<0.05).3.Cytokine test results:(1)In the state of rat healthy function: Compared with the healthy blank group,the concentration of RANTES and IL-18 in the single-acupuncture group of healthy Dazhui,Baihui and Zusanli were reduced(P<0.05),The concentration of VEGF increased(P<0.05),and the concentration of RANTES in the healthy meridian group and the healthy control group decreased(P<0.05);compared with the healthy meridian group,the concentration of RANTES and IL-18 in each healthy single-point group decreased(P<0.05),the concentration of VEGF increased(P<0.05),the concentration of RANTES in the healthy control group increased(P<0.05),and the concentration of VEGF decreased(P<0.05).(2)In the state of rat disease function: Compared with the blank group of the disease,the concentration of RANTES and IL-18 in the single-point group of acupuncture diseases Dazhui,Baihui and Zusanli decreased(P<0.05),and the concentration of VEGF increased(P<0.05),the concentration of IL-18 in the disease meridian group decreased(P<0.05);compared with the disease meridian group,the concentration of RANTES and IL-18 in each disease group decreased(P<0.05),the concentration of VEGF Increased(P<0.05),the disease control group RANTES,IL-18 concentration increased(P<0.05),VEGF concentration decreased(P<0.05).Conclusion 1.Acupuncture at Baihui,Dazhui,Zusanli single and group points can play a role in brain protection in healthy and epilepsy rats,and its mechanism may be related to reducing the rate of hippocampus positive cells and increasing nerve cell apoptosis in rats The rate is related to regulating the expression of inflammatory factors,and this effect is based on acupuncture at a single point.2.Single and group acupoints have specific targeting in regulating the early apoptosis of neurons in hippocampus tissues of rats,and are related to the body state of rats.
