Objective:The aim of this study was to identify neuromedin U(NMU)and Group 2 innate lymphoid cells(ILC2s)in Allergic Rhinitis(AR),and the possible roles of NMU and ILC2 s cells in the pathogenesis of AR were also discussed.Methods:AR patients Unilateralinferior turbinate mucosal tissues(n=10)and healthy inferior turbinate mucosal tissues(n=10)were collected,and all specimens were divided into three.One portion was used for Hematoxylin and eosin(HE)staining and immunohistochemical staining in paraffin sections to observe the histopathological status of nasal mucosa.One portion was used for immunofluorescence staining in frozen sections to analyze the expression and localization of ILC2 s and NMU in the mucosa.One portion was used for ELISA to detect the expression levels of NMU,IL-5and IL-13 in each group,and SPSS21.0 software was used for statistical analysis to compare the differences in each group.Results:1.NMU and ILC2 s were located in the nasal Mucosa of AR patients by immunofluorescence assay.2.Immunohistochemistry and immunofluorescence staining showed that the expression of NMU and ILC2 s in the nasal mucosa of AR patients was significantly higher than that of the normal control group.3.ELISA results show: Compared with the control group,the NMU expression inthe AR group increased,the difference was statistically significant[(380.85±52.52)vs(104.21±25.64),P<0.05].The results of immunohistochemistry and immunofluorescence were consistent with Elisa.Compared with the control group,the expression levels of IL-5 and IL-13 in the AR group were significantly increased,the difference was statistically significant[IL-5(23.49±3.44)vs(9.79±0.88),IL-13(34.27±3.42)vs(15.83±1.25),P<0.05].4.Here we observed that the co-localization between ILC2 s,NMU and cholinergic neurons(SNAP25+)in the nasal mucosa.Conclusion:NMU and ILC2 s are elevated in AR,and ILC2 s in the nasal mucosa co-localize with cholinergic neurons that express the NMU,it is suggested that NMU and ILC2 s cells have important significance in the pathogenesis of AR.
