Experimental Study On Protective Effect Of Glacier Quick-acting Jiuxin Dropping Pills On Myocardial Ischemia Injury

Objective: In this study,isoproterenol(ISO)was used as a predisposing factor to replicate the animal model of acute myocardial ischemia,we observed the effect of "Glacier Suxiao Jiuxin Pill" on myocardial injury in myocardial ischemic injury model animals and the mechanism of the protective effect of Jiuxin Dropping Pill on myocardial ischemic injury.Methods: Sixty healthy adult SD rats weighing 220-250 g were selected and randomly divided into blank group,model group,positive control drug group(Suxiaojiuxin Pill),Glacier Suxiao Jiuxin Dropping Pills high dose group,medium dose group and low dose group,each group has 10 rats.The blank group and the model group were given distilled water of the same volume by gavage every day.The control groupw were given gavage every day for consecutive 4 days.Starting from the 3rd day of gavage,all groups except the blank group were given ISO(4 mg/kg body weight)intraperitoneally for consecutive 2 days.The animal model of myocardial ischemia injury was reproduced by intraperitoneal injection of ISO,and then the rats under anesthesia were fixed on the fixing plate in supine position.The changes of standard II lead electrocardiogram for 5 minutes,10 minutes,20 minutes and 30 minutes in each group of animals were observed and recorded respectively.The ST segment changes of ECG were calculated,and the total millivolts(?-ST)of S-T segment changes of ECG of animals in each group was regarded as the index of myocardial injury degree,namely myocardial ischemia degree.After the observation period finished,abdominal aorta puncture was used to take blood,centrifugation was performed at 3000 rpm for 10 minutes,serum was taken and stored for later use.After we took blood,the fixed rats were operated to take out the heart tissue of the rats,and it was kept at-80? for later use.Observation index: 1.Electrocardiogram observation: we used electrophysiological techniques to observe the changes of electrocardiogram in each group,and the total millivolts'(?-ST)changes in each S-T segment was regarded as the index of myocardial injury degree.2.The changes of myocardial enzyme(LDH,CK,AST)markers of myocardial injury in each group were observed by blood biochemical detection method.3.we used Enzyme-linked immunosorbent assay to detect the changes of troponin I(c Tn I),ischemia modified albumin(IMA),malondialdehyde(MDA)and superoxide dismutase(SOD)activity in myocardial tissue homogenate in each group of animals.Results:1.the observation results of myocardial ischemia injury degree(electrophysiology)showed that the total sigma-st millivolts of rats in the high dose group,the medium dose group and the positive control group of glacier suxiaojiuxin dripping pills are lower than those in the model group at the same time,and the difference is statistically significant(P<0.05),which indicated that glacier suxiaojiuxin dripping pills can reduce myocardial ischemia injury of isoproterenol-induced myocardial ischemia model animals and have certain protective effect on myocardial injury.2.The detection results of myocardial enzymes showed that the content of Aspartate aminotransferase(AST)in serum of rats in the high dose group,medium dose group,low dose group and positive control group of Glacier Suxiao Jiuxin Dropping Pills is lower than that of the model group incompared with the model group,and the differences among the high dose group,medium dose group,low dose group and positive control group of Glacier Suxiao Jiuxin Dropping Pills are statistically significant(P<0.05),especially the low dose group has the best effect(P< 0.01);Compared with the model group,the serum CK content in the high dose group,medium dose group,low dose group and positive control group of Glacier Suxiaojiuxin Dropping Pills was lower than that in the model group,and the difference between the low dose group of Glacier Suxiaojiuxin Dropping Pills and the model group was statist ically significant(P<0.05).Compared with the model group,the serum LDH content of rats in the high dose group,medium dose group,low dose group and positive control group of Glacier Suxiaojiuxin Dropping Pills is lower than that of the model group.The difference between the low dose group of Glacier Suxiaojiuxin Dropping Pills and the model group is statistically significant(P<0.01),which can aslo suggest that Glacier Suxiaojiuxin Dropping Pills can reduce myocardial ischemia injury of isoproterenol-induced myocardial ischemia model animals and have certain protective effect.3.The detection of c Tn I and IMA in markers of myocardial injury showed that compared with the model group,the content of c Tn I in the myocardial tissue homogenate of the high dose group,medium dose group,low dose group and positive control group of Glacier Suxiao Jiuxin Dropping Pills was lower than that of the model group,and the difference among the high dose group,medium dose group and the model group was statistically significant(P<0.05),especially the high dose group had the best effect(P<0.01).Compared with the model group,the IMA content in myocardial tissue homogenate of the high dose group,medium dose group,low dose group and positive control group of Glacier Suxiaojiuxin Dropping Pills was lower than that of the model group and the differences among the high dose group,medium dose group,positive control group and model group were statistically significant(P<0.05),especially the positive control group had the best effect(P<0.01),suggesting that Glacier Suxiaojiuxin Dropping Pills can reduce myocardial cell damage of isoproterenol-induced myocardial ischemia model animals and have certain protective effect on myocardial damage.4.the detection results of MDA and SOD activity in myocardial tissue homogenate showed that compared with the model group,the MDA content in myocardial tissue homogenate of high dose group,medium dose group,low dose group and positive control group of glacier quick-acting Jiuxin dripping pill was lower than that of the model group,and the difference among the dose group,positive control group and model group is statistically significant(P<0.05),especially the positive control group had the best effect(P<0.01);Compared with model group,SOD content in myocardial tissue homogenate of high dose group,medium dose group,low dose group and positive control group of Glacier Suxiaojiuxin Dropping Pills was higher than that of model group,there were statistically significant differences among middle dose group,low dose group and positive control group of Glacier Suxiaojiuxin Dropping Pills and model group(P<0.05).Our results suggested that Glacier Suxiaojiuxin Dropping Pills can improve myocardial ischemia degree,resist free radical damage,reduce the damage caused by myocardial ischemia,and have certain protective effect.Conclusion:The experimental results showed that Glacier Suxiaojiuxin Dropping Pills can alleviate myocardial injury of isoproterenol-induced myocardial ischemia model animals to a certain extent,and its mechanism of action may be related to the mechanisms of Glacier Suxiaojiuxin Dropping Pills such as stabilizing myocardial cell model structure,resisting free radical damage,improving myocardial ischemia degree,etc.This can provide a reliable theoretical basis for the development of new drugs of Glacier Suxiao Jiuxin Dropping Pills and its clinical application in the treatment of coronary heart disease.But the detailed mechanism of its action still needs further study.In this study,isoproterenol(ISO)is used as an inducing factor to duplicate an acute myocardial ischemia animal model,to observe the effect of "Glacier Suxiao Jiuxin Dropping Pills" on myocardial injury degree of the animal model of myocardial ischemia injury,and to explore the relevant mechanism of "Glacier Suxiao Jiuxin Dropping Pills" on myocardial ischemia injury protection.
