Effects And Mechanism Of Gualou Xiebai Dropping Pills On Myocardial Ischemia Reperfusion Injury Rats

Objective: To investigate the effects of Gualou Xiebai dropping pills on anti acute myocardial ischemia in rats,and to explore the role of PI3K/Akt in acute myocardial ischemia.Therefore,provide a scientific basis for Gualou Xiebai dropping pills into clinical application.Methods: By ligating left anterior descending branch of coronary artery,the model of myocardial ischemia-reperfusion injury in rats was established and reperfused for 120 minutes after ischemia for 30 minutes.Setting up sham group,model group,Gualou Xiebai dropping pills group,compound danshen dropping pills group,wortmannin group and Gualou Xiebai dropping pills combine wartmannin group,respectively.1.With a standardⅡlead electrocardiogram(ecg)continuous monitoring to observe the change of S-T segment;2.Determination the levels of creatinine kinase,MB isoenzyme(CK-MB),myoglobin(MYO)and troponin-T(CTNT)in serum,by enzyme-linked immuno sorbent assay(ELISA)staining;3.The cardiac muscle tissue was stained by triphenyl tetrazolium chloride(TTC)to observe the areas of cardiac ischemia danger and myocardial infraction were observed;4.Using electron microscope to observe the changing of cardiac muscle cell.By HE staining,the necrosis area and pathological changes will be observing;5.By TdT-mediated dUTP Nick-End Labeling(TUNEL)kit to detect myocardial cell apoptosis;6.Use the Western blotting method testing the expression of apoptotic-related factor(caspase-3)in myocardial cells as well as the Phosphatidylinositol-3-kinase signal path gap-associated protein Akt and p-Akt.Validation the role of PI3K/Akt signal in acute myocardial ischemia induced myocardial damage occurs.Results: 1.The acute myocardial ischemia after stage of the electrocardiogram of S-T drive up can be against by Gualou Xiebai dropping pills.And compared with model group,there are obvious difference(P<0.01).However,there are no statistical significance compared with compound danshen dropping pills.2.Compared with sham group,model group of serum CK-MB,MYO and CTNT content increased significantly(P<0.01 or P<0.05);Gualou Xiebai dropping pills group can obviously reduce serum CK-MB,MYO and CTNT content in rats,but there are no statistical significance compared with compound danshen dropping pills.3.Compared with model group,Gualou Xiebai dropping pills group can distinctly reduce infarction percentage significantly,and this effection close to compound danshen dropping pills.4.Gualou Xiebai dropping pills group can decrease the condition of myocardial cell fibers arranged disorder,decrease between the muscle fiber inflammation cells infiltration,myocardial cell degeneration and necrosis,and decrease myocardial cell interstitial edema,according to the electron microscope and light microscope results.5.According to the TUNEL results,Gualou Xiebai dropping pills group can effectively inhibit the apoptotic of myocardial cell,and the same with compound danshen dropping pills.6.Western blot results show that,Gualou Xiebai dropping pills group can effectively inhibit the expression of caspase3 and improve the protein of p-Akt in acute myocardial ischemia rats,and these demonstrating that one of the mechanisms associated with the activation of PI3K/Akt signaling pathways.Conclusion: Gualou Xiebai dropping pills resistance to the effect of the myocardial injury caused by myocardial ischemia is obviously,as well as compound danshen dropping pills.Although the result of Gualou Xiebai dropping pills against the electrocardiogram of ST drive up same with compound danshen dropping pills,Gualou Xiebai dropping pills have more apparent confrontation effect about the fall back of ST segment.In addition,Gualou Xiebai dropping pills can reduce the content of CK-MB、MYO、CTNT in serum;Obviously narrow the myocardial infarction scope;decrease the number of myocardial cell apoptotic and safeguard myocardial tissue;PI3K/Akt signal path have played a very important role in the process of Gualou Xiebai dropping pills resistance to the effect of the myocardial injury caused by myocardial ischemia.
