Objective:This thesis examined the expression and clinical significance of PRRX1,Smad2,Vimentin,and E-cadherin in gastric cancer tissues and normal tissues adjacent to the cancer.To investigate the role of PRRX1 and Smad2 in epithelial-mesenchymal transition of gastric cancer and the effect of prognosis.Methods : 86 gastric cancer tissues and normal adjacent tissues were collected and the expressions of PRRX1,Smad2,E-cadherin and Vimentin were detected by immunohistochemical SP method.Spearman's method was used to analyze the among PRRX1 and Smad2,Vimentin,and E-cadherin.The relationship between PRRX1,Smad2,Vimentin,and E-cadherin expression survival in gastric cancer patients were estimated and analyzed by KaplanMeier method.Results : The positive expression rates of PRRX1,Smad2,and Vimentin were higher in gastric cancer tissues than in normal adjacent tissues(P <0.05).However,the positive expression rate of E-cadherin in gastric cancer tissues was lower than that in normal adjacent tissues(P <0.05).Gastric cancer had poor histological differentiation(G3),deep tumor invasion(T3-T4),number of metastatic lymph nodes> 5 and PG??70ng / ml,and the expression levels of PRRX1,Smad2 and Vimentin were significantly increased(P <0.05).E-cadherin expression was significantly decreased in gastric cancers with more than 5metastatic lymph nodes and late TNM stage(?-?)(P <0.05).PRRX1 was positively correlated with Smad2 and Vimentin expression(r values were 0.444,0.278,P <0.05),but negatively correlated with E-cadherin expression(r =-0.312,P <0.05).The median survival times of PRRX1 positive and negative expression were 26 months(95% CI: 23~29 months)and 35 months(95% CI: 33~37 months),and the difference wasstatistically significant(P=0.040).The median survival times of Smad2 positive and negative expression were 27 months(95% CI: 22 ~ 32 months)and 34 months(95% CI: 31 ~ 37months),and the difference was statistically significant(P=0.008).The median survival times of Vimentin positive and negative expression were 26 months(95% CI: 24~28 months)and36 months(95% CI: 33 ~ 39 months),and the difference was statistically significant(P=0.003).The median survival times of E-cadherin positive and negative expression were 36months(95% CI: 34 ~ 38 months)and 26 months(95% CI: 24 ~ 28 months),and the difference was statistically significant(P=0.000).Conclusion:Gastric cancer is positive for PRRX1,Smad2,and Vimentin,and which were associated with poor prognosis in patients.PRRX1 expression was positively correlated with Smad2 and Vimentin,but negatively correlated with E-cadherin.PRRX1 and smad2 may participate in the epithelial-mesenchymal transition process of gastric cancer,which may be a potential target for early intervention of gastric cancer recurrence and metastasis.
