Objective To investigated the expression and prognostic value of nuclear transcription factor ?B(NF-?B),Glutamate-cysteine ligase catalytic subunit(GCLC)and Glutamate-cysteine ligase regulatory subunit(GCLM)in T-cell lymphoma(TCL)tissues.Methods 1.Fifty-eight paraffin-embedded specimens of TCL patients who were initially diagnosed by pathological biopsy in Gansu Provincial Hospital from January 2013 to December 2018 were collected,mainly including 19 cases of extra-nodal NK/T-cell lymphoma,nasal type(ENKTL),9 cases of angioimmunoblastic lymphadenopathy(AITL),12 cases of peripheral T-cell lymphoma,not otherwise specified(PTCL-NOS),6 cases of anaplastic large cell lymphoma,ALK-positive(ALK+ ALCL),7 cases of anaplastic large cell lymphoma,ALK-negative(ALK-ALCL),2 cases of enteropathy type T cell lymphoma(EATL),3 cases of subcutaneous panniculitis like T cell lymphoma(SPTCL)and 1cases of Mycosic Fungoides(MF).Meanwhile,20 cases of reactive hyperplastic lymphoid tissue were collected as control group.2.The levels of NF-?B in 58 TCL tissue samples and20 cases of reactive hyperplastic lymphoid tissue were detected by immunohistochemistry,and we also analyzed the correlation between the expression of NF-?B and TCL clinicopathological parameters.3.The immunohistochemistry was also performed to detect expressions of GCLC and GCLM in TCL tissue and reactive hyperplastic lymphoid tissue,and the relationship between their expressions and the clinicopathological characteristics ofTCL patients was further analyzed.4.To analyze the correlation between the expression GCLC and GCLM in TCL tissue.Results The positive rate of NF-?B in TCL tumor tissue was 75.86%,which was higher than that in lymph node reactive hyperplasia tissue(35.0%)(P<0.05).The expression of NF-?B was significantly correlated with Ann-Arbor clinical stage,B symptoms,and international prognostic index(IPI),but was not affected by age,gender,LDH level,or pathological typing(P >0.05).The positive expression rates of GCLC and GCLM in TCL tissues were higher than that in the lymph node reactive hyperplasia tissues(P<0.05),and the expression of them was correlated with B symptoms,Ann-Arbor clinical stage and international prognostic index(IPI),but not associated with age,gender,LDH level or pathological types(P> 0.05).Furthermore,Spearman rank correlation analysis showed that the expression of GCLC protein was positively correlated with the expression of GCLM protein in TCL tissues(r=0.376,P=0.004).Conclusion 1.The expression of NF-?B in TCL tissues is higher than that in reactive proliferative tissues of lymph nodes,and is closely related to tumor stage,B symptom and IPI,which may participate in the occurrence,development and invasion and metastasis of TCL,and is expected to become a therapeutic target of TCL.2.The levels of GCLC and GCLM in TCL tissues were higher than those in reactive lymph node hyperplasia tissues.3.The expression of GCLC protein was positively correlated with the expression of GCLM protein in TCL tissues,the interaction of them may provide theoretical basis for the etiology and treatment of TCL.
