Objective:To establish a stable red ginseng processing technology to provide high quality and stable red ginseng raw materials for Shenmai injection.Method:1.Study on the processing technology of red ginseng?1?The orthogonal test method was used to investigate the factors such as infiltration,steaming time,drying temperature and drying time.The infiltration effect and the total saponin content were used as indicators to determine the red ginseng processing technology.?2?High performance liquid chromatography was used to compare the content of Rg1,Re and Rb1 monomer saponins of homemade red ginseng and commercially available red ginseng.?3?Compare the content of red ginseng protein by Bradford method.2.Protective effect of red ginseng,a raw material of Shenmai injection,on oxidative damage of myocardial cellsHydrogen peroxide?H2O2?induced H9c2 rat cardiomyocytes to establish an oxidative stress injury model of cardiomyocytes in vitro;MTT assay for cell viability;kit for detection of cytokine LDH release,SOD activity and MDA content;flow cytometry for ROS detection,Apoptosis;Western Blot assay for apoptosis-related protein expression.Results:1.The process of red ginseng was 3d,the ginseng was 20min,the steaming time was 3h,the drying temperature was 60°C,and the drying time was 10h.2.After the processing,the red is involved in the commercial red ginseng comparison,the saponin content is stable and uniform,and the protein content is relatively low.3.Compared with the Control group,H2O2 can significantly reduce the cell survival rate.After pretreatment with red ginseng active ingredients,the survival rate of cardiomyocytes is significantly improved,and the oxidative damage of rat cardiomyocytes induced by H2O2is alleviated.4.Compared with the Model group,red ginseng significantly increased the SOD activity of H2O2-induced cardiomyocytes,inhibited the increase of MDA levels,and decreased ROS levels.5.Compared with the Model group,the pro-apoptotic protein Bax was down-regulated,the anti-apoptotic protein Bcl-2 was up-regulated,and the Bcl-2/Bax ratio was increased,which indicated that the apoptosis of cardiomyocytes was alleviated by alleviating the expression of apoptosis-related proteins..Conclusion:The processing technology of red ginseng suitable for Shenmai injection was established to ensure the uniform quality of red ginseng products.It was found that red ginseng,a raw material of Shenmai injection,had a good protective effect on H2O2-induced oxidative stress injury in H9c2 rat cardiomyocytes.
