Diarrhea is defined as a disease of three or more excretions a day with increased water content and thinned stools.Diarrhea is mostly diagnosed in children under 5years old,as a consequence,kids would have dehydration and even death.Therefore,the treatment of diarrhea is a significance study.In this paper,we focused on the antidiarrheal effect and toxicity of the infant warm stomach tea,which is composed of pine nuts,gardenia,black tea and so on.The study was conducted as the following methods:In the first part,the infant warm stomach tea was extracted with water in the rate of 2.43%.Secondly,its the antidiarrhea experiment was carried out,and the diarrhea model of mice was established by senna leaf.The appearance signs and diarrhea of mice were observed and caculated,and the diarrhea evaluation indexes such as defecation inhibition rate,diarrhea inhibition rate,loose stool rate and diarrhea index were calculated.The effects of the infant warm stomach tea on the small intestine propulsion and the small intestine effusion of rats were observed by using the ink propulsion experiment and the small intestine effusion experiment of rats.Finally,acute toxicity test was carried out in mice at 25 times of the highest dose(290mg/kg).Long-term toxicity test on 80 rats,half male and half female.The drug was daily taken for 14 days and stopped for 7 days.The physical signs,blood routine,organ coefficient,blood biochemical indexes and tissue pathological were observed.In the second part,the effect of water extract of pine nut shell on diarrhea model induced by castor oil was studied.The external signs and diarrhea of mice were observed and recorded.The diarrhea delay rate,defecation inhibition rate,diarrhea inhibition rate and diarrhea index in vivo were calculated.The polysaccharide was extracted from pine nut shell by water and precipitated by ethanol,deproteinized by sevec and papain,and purified by resin decolorization,dialysis and hierarchical ethanol precipitation.The structure was characterized by UV,IR,GPC and GC-Ms.The antibacterial activity of Polysaccharide from pine nut shell was studied by plate counting method.The results are as follows:(1)compared with the model group,the number of stools in the positive group and the administration group decreased significantly(P<0.01).Compared with the model group,the number of loose stools in each group decreased significantly(P<0.05).The diarrhea inhibition rate of the positive group was 20.00%,and the high dose group and the middle dose group was 22.22%and30.00%,higher than that of the positive group.(2)Compared with the untested group,the small intestine peristalsis index of the low,middle and high dose group and the positive control group decreased significantly(P<0.05),and the propulsion inhibition rate was 16.2%,33.7%,17.1%and 24.1%,respectively.(3)Compared with the model group,the volume of small intestinal effusion in the low,middle and high dose group and the positive control group decreased significantly,and the inhibition rates of small intestinal effusion were 16.11%,21.52%,25.20%and 26.08%,respectively.(4)According to the acute toxicity and long-term toxicity experiments,there was no poisoning and death.(5)In the experiment of castor oil induced diarrhea,compared with the model group,the water extract of pine nut shell prolonged the first diarrhea time significantly(P<0.05),and the defecation inhibition rate and diarrhea inhibition rate were similar to those of the positive group.(6)The polysaccharide with molecular weight of 2.6×10~4 Da was obtained from pine nut shell,which was composed of galactose,mannose,glucose,arabinose,xylose and rhamnose.The molar percentage was 30.5:28.6:19.3:12.4:5.2:3.9.The polysaccharide of pine nut shell mainly consists of galactose residue?4)-GALP-(1?and mannose residue?4,6)-manp-(1?,man-(1?,?3,6)-manp-(1)?.The results of plate counting showed that the polysaccharide of pineal shell had antibacterial activity against S.aureus and showed concentration dependence.Conclusion:the infant warm stomach tea show the effect of antidiarrhea and no toxicity.The water extract of pine nut shell also has a certain antidiarrhea effect.The natural polysaccharide with uniform molecular weight purified from pine nut shell has the activity of inhibiting Staphylococcus aureus.
