Objective To investigate the effect of overexpression of odd-skipped related transciption factor 1(OSR1)on the proliferation,migration,invasion and cell cycle arrest of HCT-8、LoVo colon cancer cells.Methods The recombinant lentivirus vectors pEZ-Lv105-OSR1 and blank vector pEZ-Lv105 were transfected into HCT-8、LoVo colon cancer cells,respectively,the protein levels of OSR1 were detected by Western blot.The abilities of proliferation,metastasis,invasion and cell cycle arrest of HCT-8、LoVo colon cancer cells were detected for the effects of OSR1 overexpression.The protein expression levels of E-cadherin,N-cadherin,Vimentin,p-NF-KB p65,NF-KB p65 and p-IκBα were detected by Western blot.Results The expression level of OSR1 protein was significantly lower in pEZ-Lv105-OSR1 than in pEZ-Lv105 control group(P<0.05).The pEZ-Lv105-OSR1 transfected group had markedly inhibited abilities of colon cancer cells proliferation,migration,invasion and induced coloncancer cells cell cycle arrest compared to the control group(P<0.01).The protein expression level of E-cadherin was higher in transfected group than in control group(P<0.05),but N-cadherin,Vimentin,p-NF-KB and p-IκBα expression levels were lower in pEZ-Lv105-OSR1 transfected group than in pEZ-Lv105 control group(P<0.05,P<0.01).The expression of NF-KB p65 was unchanged.Conclusion Overexpression of OSR1 inhibits the abilities of proliferation,metastasis,invasion and cell cycle arrest of HCT-8 、LoVo colon cancer cells by NF-KB signal pathway. |
