Deubiquitinating enzymes (DUBs) are a class of proteases which can reverse the ubiquitination. Recent studies have found that deubiquitination enzymes not only play an important role in the normal physiological activity, but also may correlations with the tumorigenesis and development. OTU domain-containing ubiquitin aldehyde-binding protein1(OTU1) is a number of deubiquitinating enzymes family, but its exact molecular mechanism in tumor is still unclear. So we have explored OTU1in colon cancer, and then understand the relationship between OTU1and human tumors. Real-time PCR and immunohistochemistry were carried out in selected colon cancer samples. The cell line which was overexpression of OTU1was established by liposome. However, the cell line which was specific interference of OTU1was established by lentiviral vector system. CCK-8test was used to analyze the growth of colon cancer cells after overexpression of OTU1and interference of OTU1. And then, Transwell and Matrigel methods were used to analyze the migration and invasion abilities of colon cancer cells. The experiments results showed that:the expression of OTU1mRNA was3.15-fold higher in colon cancer than that in adjacent normal mucosa (P<0.05); and the expression of OTU1protein in the colon cancer was significantly higher than the adjacent colonic mucosa (P<0.05). The higher expression of OTU1in colon cancer was related with tumor size, differentiation and metastasis of lymph node (P<0.01). High expression rates of OTU1in adjacent normal mucosa, adenoma and carcinoma were11.11%,55.56%and77.78%respectively (P<0.05). Overexpression of OTU1can promote cell proliferation in vitro, promote cell migration and invasion. On the contrary, interference of OTU1can inhibit cell growth, migration and invasion. In conclusion, the expression level of OTU1mRNA and protein in colon cancer tissues were significantly increased. OTU1can promote cell proliferation, migration and invasion of colon cancer. OTU1plays an important role in tumorigenesis, development and metastasis of colon cancer.
