ObjectiveThe current study is aimed to investigate the potential role of BAG3 in regulating proliferation,migration and invasion of colon cancer HCT-116 cells.This study provides a theoretical basis for the development of new anticancer drugs by finding colon cancer-related drug targets.Methods1.Immunohistochemistry was used to detect the expression of BAG3 in colon cancer clinical samples.2.Stable BAG3-overexpression HCT-116 subline was generated by lentivirus infection.The expression of BAG3 was measured by RT-PCR and Western blot.3.BAG3-knockdown HCT-116 subline was generated by CRISPR/CAS9 lentivirus infection.The expression of BAG3 was measured by PCR,DNA agarose gel electrophoresis,Surveyor and Western blot.4.Cell proliferation was investigated by cell counting,cell colony formation,Ed U and real-time cell analyzer(RTCA).5.Migration and invasion was investigated by wound healing and Transwell.Results1.The expression of BAG3 in colon cancer tissues was higher than in normal tissues.2.Stable BAG3-overexpression HCT-116 subline and BAG3-knockdown HCT-116 subline were successfully constructed.3.In colon cancer HCT-116 cells,overexpression of BAG3 promotes proliferation,knockdown of BAG3 inhibits cell proliferation.4.In colon cancer HCT-116 cells,overexpression of BAG3 promotes migration and invasion,knockdown of BAG3 inhibits migration and invasion.ConclusionsBAG3 plays an important role in promoting cell proliferation,migration and invasion of colon cancer cells HCT-116.
