Objective:To detect the expression of neurogenin3(NEUROG3)in gastric cancer tissues and cells,and its relationship between the degree of gastric cell differentiation and clinicopathologic stage.Methods:The clinicopathological data of 181 patients in The Second Affiliated Hospital of Chongqing Medical University during 2016 to 2018 were retrospectively collected.Immunohistochemical(IHC)staining were used to detect the expression of NEUROG3 in intestinal metaplasia gastric mucosa and gastric cancer tissues.The expression of NEUROG3 in gastric cancer cell lines with different differentiation and GES-1 was detected by Real-time PCR and Western blot.The morphological changes of MKN-28 cells with NEUROG3 gene silencing were observed,and the expression of inhibitor of DNA binding or differentiation(id-1)was detected by real-time PCR.Results : NEUROG3 expression was lower in low-differentiated gastric cancer tissues,but higher in paracancer tissues,intestinal metaplasia gastric mucosa tissues and medium-high differentiated gastric cancer tissues(P<0.001).The expression of NEUROG3 in GES-1 and MKN-28 was significantly higher than that in SGC-7901,MKN-45 and BGC-823.After NEUROG3 was silenced in MKN-28 cells,the degree of cell differentiation was decreased,and the expression of Id-1 was significantlyincreased(P<0.001).Furthermore,the clinical association between NEUROG3 and gastric cancer was evaluated.There were no statistical significance between NEUROG3 expression in gastric cancer tissue and patients' gender,age or tumor size(P?0.05).However,NEUROG3 expression was correlated with the degree of tumor differentiation(P<0.001,r= 0.412),lymph node metastasis(P<0.05,r =-0.180)and TNM stage(P<0.001,r=-0.431).Conclusion : The expression of NEUROG3 gene was positively correlated with the degree of gastric cancer differentiation and negatively correlated with the clinical and pathological stage.
