Garlic can be used as food or condiment as well as medicinal material.It has a long history in China.As early as in the "Compendium of Materia Medica",it is recorded that garlic "passes the five internal organs,reaches various tactics,removes cold and dampness,avoids evil,and eliminates bloatedness." Modern studies have shown that garlic has the functions of eliminating tumors,lowering blood pressure,lowering blood lipids,lowering blood sugar,preventing blood clots and protecting the liver,in addition to weight loss,beauty,and smoking cessation.Domestic studies have shown that both garlic polysaccharides and alliin have liver-protective effects,but the selected indicators for evaluating garlic liver damage are too simple.In this study,garlic extract was prepared first,and then the garlic extract was used to study its protective effect on carbon tetrachloride-induced liver injury in mice.The specific research contents are as follows: 1.Preparation of garlic extract and determination of alliin contentOrthogonal experiment was used to study the effect of material-liquid ratio,reaction time and extraction method on the content of alliin in garlic extract,and a high-performance liquid chromatography method for quantifying alliin was established.The content of alliin in garlic in Sichuan,Yunnan and Henan.The result is that the effect of the content of alliin in garlic extract is the ratio of material to liquid> extraction method> reaction time.The optimal conditions for extraction are 1:4 liquid-to-liquid ratio,stirring for 10 minutes,chromatographic conditions using a C18 column,mobile phase 10% methanol 90% water,flow rate 1mL/min,time 40 min.The content of alliin in Shandong,Yunnan,and Sichuan all exceeds 1%.Among them,garlic in Shandong has the highest content of alliin.Garlic in these areas can be used as the raw material for alliin extraction.2.Protective effect of garlic extract on carbon tetrachloride-induced liver injury in mice76 6-week-old SPF-free ICR mice were randomly divided into 6 groups.The negative control group control group and the model group were administrated with pure water.The garlic extract was administrated with garlic extract at the low,medium,and high dose groups.The control group was given intragastrically.After continuous administration for 14 days,after the last administration,the model group and each administration group were perfused with carbon tetrachloride edible oil solution to prepare a mouse liver injury model,and the negative control group was perfused with edible oil.Detection of total protein(TP),albumin(ALB),glutathione(GSH)in serum and liver of mice and Aspartate Aminotransferase(AST)in mouse serum and liver,Alanine Aminotransferase(ALT),Lactate Dehydrogenase(LDH),Choline esterase(CHE)activity in the liver.The results showed that garlic extract could inhibit the abnormal increase of AST,ALT,LDH activity in mice caused by carbon tetrachloride,and inhibit the abnormal decrease of TP,ALB,GSH content in mice caused by carbon tetrachloride.It is speculated that by increasing the concentration of GSH to remove oxygen free radicals in liver cells and improve the stability of liver cell membranes,garlic extract can reduce the degree of liver damage and bring the level of metabolism of liver cells close to normal levels.
