Objective: Garlic (Allium sativum L.) is a Liliaceous Allium perennial herbaceousplants which is recognized as a medicine and food treasures. Garlic is one of the hottopics in the study of modern medicine. Alliin is the main efficacy component and couldbe catalysd by enzyme of alliinase to allicin. A little research on alliin’s quality,especially the disposal in vivo. This study will build QTOFMS method analysis two kindsof alliin pilot products. Explore the different quality of alliin with near infrared spectrum.The NIR will provide a quick analysis method for the subsequent production processquality control. In order to understand the disposal of alliin in blood and other biologicalsamples, this study will also explore SPE method of alliin in blood sample and establishrelevant methodology. Methods: Using QTOFMS analysis for alliin garlic extractingredients. Adopting the method of near infrared for alliin extract and50%of differentbatches of refined alliin. Using SPE method for extraction and separation of alliin inbiological samples, and establish the UPLC-MS/MS method for the determination ofalliin content. Results: The purity of different alliin garlic extract results show that90%of alliin besides have alliin, otherwise to alliin dipole ions formation of dimer, trimer andfour polymers. There are also arginine and proline dimer dipole ions in90%alliin In50%of alliin, in addition to alliin dipole ions formation of dimer, trimer, glutamine, proline,arginine and proline dimer dipole ions, methyl alliin and garlic amino acid sodium salt,potassium dimer dipole ions, deoxidization alliin and garlic amino acid sodium salt,potassium dimer dipole ions are also found. Analysis alliin extracts of different puritywith near infrared spectrum, the differences are found. The qualitative model of alliinextract (50%) was established. Alliin in blood samples was analysis by cation exchangecolumn solid phase extraction. On the basis of positive pressure recovery more than65%,UPLC-MS-MS determination of alliin in plasma of rats and intestinal absorptionsolution of linear range are1.52μg/ml~30.30μg/ml. The precision and recovery wereconform to the requirements of the biological sample analysis method validation. The method eastablished was basically eliminate the matrix effect. Conclusion: This studyprovide the elementary data for the future material pilot production and new drug research.The near infrared method for alliin raw materials and preparation process of productionprovide a quickly and reliability analysis. SPE-UPLC-MS/MS settled a methodologyfor the future research of alliin dynamics characteristics, absorption, transport andmetabolism in vivo.
