Objective To explore the anti-tumor effect of the polyphyllin ?,a component of the Zhuang medicine Kezhonglou,and its effect on the immune system of tumor-bearing mice.To study the role of polyphyllin I in the IKBKE/NF-?B pathway.Provide scientific basis and theoretical guidance for exploring the anti-cancer medicinal value of polyphyllin ?.Methods The experiment is divided into animal experiment and in vitro experiment.1.Balb/c mice inoculated with 4T1 cells under the arm to induce breast cancer tumor models,randomly divided into normal group,breast cancer model group,PPI high dose group(1mg/kg),PPI low dose group(0.5mg/kg)And paclitaxel group(1mg/kg).Intraperitoneal injection was given for 14 days.Serum COX-2,IL-2,IL-6 levels were measured by ELISA,and the spleen and thymus indexes of mice were calculated.The contents of IKBKE,AKT and NF-?B in tumor tissue were measured by immunohistochemistry.2.Carry out CCK-8 experiment and cell scratch experiment using 4T1 mouse breast cancer cells,and determine the inhibitory effect of different concentrations of saponin ? on the growth and migration of 4T1 cells.Flow cytometry was used to measure cell cycle and apoptosis.Cellular protein was extracted and the expression of IKBKE,AKT,NF-?Bp65 in 4T1 cells was detected by Western blot and PCR technology.Results 1.Polyphyllin ? can down-regulate the serum COX-2,IL-2,IL-6 levels in tumor-bearing mice,which is significantly different from the model group.Compared with the normal control group,the thymus index and spleen index of tumor-bearing mice decreased,and the thymus index and spleen index of the administration group were higher than the model group.2.The results of CCK-8 and cell scratching experiments showed that the effect of polyphyllin I on inhibiting the growth and metastasis of 4T1 cells increased with increasing concentration.The results of flow cytometry showed that polyphyllin I blocked breast cancer cell cycle at g0/g1 phase and promoted apoptosis.The results of Western blot and PCR showed that polyphyllin I can inhibit the expression of IKBKE,AKT and NF-?B in 4T1 cells.Conclusion Polyphyllin I can reduce the content of serum inflammatory factors COX-2,IL-2,IL-6,increase the thymus index and spleen index of breast cancer tumor-bearing mice,and reduce the content of IKBKE and NF-?B in the tumor tissue of mice.The immune system has a protective effect.Polyphyllin I can inhibit the growth and metastasis of 4T1 breast cancer cells,block the cell cycle,inhibit the expression of IKBKE,AKT,NF-?B,induce apoptosis and play an anti-tumor effect.
