Objective: Breast cancer is a malignant tumor which is serious threat to women’s health. It is the leading cause of death in various pathological types of breast, and the incidence is growing. Now, breast cancer has become the most common malignant tumor in women, and presents a new development to the trend of younger age, in most pathological types of breast, the smaller the sick patients are, the higher the invasiveness is, and the overall prognosis is worse. Breast tissue is rich in blood vessels, so is prone to metastases. Neovascular provides nutrients for breast cancer, removes metabolic products of breast cancer and can also lead to the development of metastasis. Therefore, angiogenesis plays an important role in the occurrence and development of breast cancer. In recent years, anti-angiogenesis therapy in tumor is becoming a hot topic in scholars’ study. In this study we observed the effect of tamoxifen on nude mouse transplanted tumor, the changes of the CD31、 VEGF index in tumor tissue in nude mice after treating with tamoxifen and the influence of tamoxifen on VEGF in serum of tumor bearing nude mice. According to these, it provides the experimental basis of anti- angiogenesis therapy of tamoxifen for breast cancer. Methods: Fourteen BALB / C- Nu female nude mice, 4~6 weeks, human breast cancer SK-BR-3 cells were inoculated in nude mice right forelimb armpit in the second mammary fat pad. And it was randomly divided into 2 groups, control group with 7 rats, tamoxifen group with 7 rats. According to the dosing regimen respectively to mice in 2 groups to intragastric administration every-day. Recode the general living conditions in nude mice, Dynamic observation of the growth in nude mice and change of its weight Measure. The xenograft tumor volume and draw the growth curves. After 14 days, nude mice was killed, weighing the tumor weight, tumor inhibition rate was calculated. The level of VEGF in serum of tumor bearing nude mice was detected by ELSA, platelet endothelial cell adhesion moleculesn(CD31) in tumor tissue and vascular endothelial growth factor(VEGF) expression were detected by immunohistochemistry method.All the experimental data were analyzed with SPSS19.0, statistical treatment of experimental results are indicated with the standard deviation, using two samples t test on the relevant experimental data analysis. With P<0.05 for the difference had statistical significance.Result: ①We proved that there is no significant effect of tamoxifen on nude mice eating,weight and so on.②With the change of time, Nude mice tumor volume in the tamoxifen group was less than control group. The difference was statistically significant(P<0.05).Tamoxifen obviously inhibitted the growth of subcutaneous transplanted tumor volume.③The inhibit rate of Tamoxifen group was 43%; the average weight of transplantation tumor were compared between the two groups, the difference was statistically significant(P< 0.05).Tamoxifen has growth inhibition in tumor.④VEGF levels in the serum of control group of tumor bearing nude mice was significantly higher than that in tamoxifen group. Statistically was significant differences(P< 0.05). Tamoxifen obviously inhibited the level of VEGF in serum.⑤The result of immunohistochemistry showed the expression of VEGF and CD31 in control group of nude mice with were significantly higher than the expression in the tamoxifen group. Statistically was significant differences(P<0.05). Tamoxifen can reduce the expression of VEGF and CD31.Conclusions: Through the establishment of SK-BR-3 human breast cancer in nude mice model, the application of tamoxifen inhibits the secretion of VEGF in nude mice serum, reduces the expression of VEGF and CD31 in the tumor tissues, can effectively inhibit the growth of transplantation tumor in nude mice. Thus for breast cancer treatment of anti-angiogenesis therapy provides a new way of thinking.
