| Aesculetin as one of the main active ingredient in traditional Chinese medicine Cortex Fraxini, which has anti-tumor activity and many other pharmacological functions. These days, did not see aesculetin contact between the body’s immune function at home and abroad.Studies have shown that aesculetin has anti-tumor function in vivo and can inhibit the growth or induce apoptosis of tumor cells. Aesculetin whether can through comprehensive regulation of tumor immune systems to enhance immune function to further exert its anti-tumor function? In this study, an aesculetin as experimental materials in the basic of immunology theory, using of experimental techniques and methods in vivo, through the research of aesculetin tumor-bearing mouse immune system, to further revealed its anti-tumor function and effect mechanisms, to provide the basis for better development and use of traditional Chinese Qin skin.Objective:to investigated the effect of aesculetin on S180tumor immune function in mice, studied and provided experimental evidence for the clinical application of the mechanism.Methods:(1) study the inhibition of AES in S180tumor in mice.(2) Studied the effect of AES on immune organs in S180mice.(3) Research AES on S180tumor mouse peritoneal macrophages activity by MTT.(4) Studied on effect of AES on the mouse spleen cells in S180antibody formation using UV spectrophotometer.(5) Studied on the effect of AES on lymphocyte transformation in S180by MTT.(6) Studied on the effect of AES on the killing activity of mice with S180tumor cytotoxic T lymphocyte (CTL) by MTT.(7) Studied on the effect of AES on the killing activity of NK of mice with S180tumor by MTT.(8) Using enzyme-linked immunosorbent assay(ELISA) to detected the effect of AES on the S180tumor-bearing mice peripheral blood serum IFN-y and IL-2.Results:In each group, Aesculetin can increase the spleen and thymus of mice and there was a significant difference compared with the negative group, indicating aesculetin has the promotion on S180tumor immune organ function; Aesculetin can promote lymphocyte transformation in S180. there is a significant difference compared with the negative group; Aesculetin dose-dependently enhanced CTL killing activity of tumor-bearing mice cells and NK cells and increased serum cytokines content of IFN-γ and IL-2.Conclusion:Aesculetin in vivo had anti-tumor activity, it can enhance non-specific immune function in mice, improve humoral immune function and enhance humoral immune function activity. Meanwhile aesculetin can enhance immune cell function in mice with some dose-dependent. Aesculetin can enhance the immune function of S180in mice in order to exert its anti-tumor effect. |
