PurposesUveitis is a common blinding inflammatory disease in the eye,often caused by autoimmune disorders.Previous studies have found that berberine can relieve clinical symptoms of autoimmune diseases such as experimental autoimmune encephalomyelitis(EAE)and ulcerative colitis.The purpose of this experiment is to investigate whether berberine can inhibit the development of experimental autoimmune uveitis(EAU)and clarify its possible role and mechanism.MethodsB10R.? mice were induced to EAU by immunization with inter-photoreceptor retinoid-binding protein(IRBP161-180).And on the first day of induction,berberine or ddH2 O was given by gavage.On the14 th day after immunization,observe the changes of the anterior chamber of the inflammatory mice with a slit lamp and perform clinical scoring;take the mouse eyeballs for HE section and perform pathological scoring;the Evans blue experiment was used to observe the changes in the integrity of the blood-retinal barrier;RT-PCR is used to detect the expression ofIL-17,IFN-r and IL-10;flow cytometry is used to detect the change in the ratio of Th1 cells,Th17 cells and Treg cells;transcriptome sequencing is used to detect the spleen transcriptome differentially expressed genes;stool16sRNA sequencing is used to detect differences in gut microbiota.ResultsOn the 14 th day after EAU modeling,compared with the control group(ddH2O group),the clinical score and pathological score of the berberine treatment group were significantly reduced,and the damaged blood-retina barrier was repaired.The expression of inflammatory factors in the berberine treatment group was significantly reduced,while the expression of anti-inflammatory factors was significantly increased.In addition,the spleen transcription group and gut microbiota of the berberine-treated group were significantly different from the control group.Conclusions1.Berberine can significantly reduce the clinical and pathological scores and blood-retina barrier function of the EAU model;2.Berberine can regulate the balance of effector T cells and regulatory T cells;3.Berberine can significantly change the spleen transcriptome of EAU mice;4.Berberine can significantly change the gut microbiota of EAU mice;5.Gut microbiota plays an important role in berberine treatment of EAU;...
