Objectives:To observe the effect of the new anti-uveitis medicinal preparation XZ on experimental autoimmune uneitis(EAU) and its mechanism of inhibiting inflammation action. Methods:The retinal S antigen emulsified in complete Freund's adjuvant injected in foot pads, hindlimbs and subcutaneous in back of the lewis rats on day1 and day7, then,salmonella typhimurium lipopolysaccharide injected in vitreous to establish the improved animal model of EAU on day8. The EAU rats were randomly divided into three group:normal control group, model control group, XZ treatment group(6mg/one,12mg/one) and dexamethasone treatment group(10mg/kg) as a positive control, 8 rats in each group. Through day9 to day20, the XZ in various dosage and dexamethasone were regularly given to the rats by oral administration once a day. Symptoms of EUA rats were observed by slit lamp microscope every three days. At Day 10, 13, 16, 19 left eye aqueous humor was gathering for ELISA,which was employed to calculate the content of IL-6 and TNF-? in aqueous humor. At day12 and day20, we make paraffin slices of rat eyeball exemplar to observe the pathological manifestations and detect IFN-? and IL-17 expression in ocular tissues by immunohistochemistry. Results:1.Clinical score: the normal rats did not see obvious inflammatory performance, while the model rats began to appear inflammation for the first time on day7, reach the peak on day10, then gradually fade from day16. The clinical score of XZ and dexamethasone treatment group was lower than that in control group, the difference was statistically significant(p<0.05).2. Histopathology: the retina structure of the normal group is clear and no infiltration of inflammatory cells. Compared with the model group, which is charactered by a large number of inflammatory infiltrates anterior chamber and each layer of retina structure badly disordered, the eye tissue lesion severity of each treatment group was significantly reduced. 3. Immunohistochemical: We detect IFN-? and IL-17 expression in ocular tissues of the model group. IFN-? and IL-17 expressed significantly high in the peak period of inflammation, while the expression in each treatment group rats decreased obviously(p<0.05).4. ELISA: in the peak of inflammation period, the model group shows that TNF-a and IL-6 in the aqueous humor increased significantly, compared with the normal group, while it gradually decreased in the recovery period. At various time points, TNF-a and IL-6 levels in the aqueous humor of each treatment group were significantly lower than the model group(p<0.05), Conclusions:Utilizing retinal s antigen combined with salmonella typhimurium lipopolysaccharide can successfully establish experimental uveitis model in Lewis rats. The new anti-uveitis medicinal preparation XZ could ameliorate EAU, down-regulate the expression of Th1 and Th17 cell related factors.
