Ginseng has a wide range of regulatory effects on the human body,and its main functional components include ginsenosides,ginseng polysaccharides,and ginseng polypeptides.Under the action of intestinal flora and digestive enzymes,the main ginsenosides are converted into rare ginsenosides,which are absorbed into the blood system and exert their effects.In view of the large differences in intestinal flora,the unhealthy people's intestinal flora is often in an unbalanced state,the content of polysaccharides and polysaccharides in natural ginseng is very low,and the individual differences are very large,which makes it difficult for ginseng to take full effect.Based on the above problems,this paper discusses the technology of human participation in enzymes,microorganisms,and enzyme-microbial coupling treatment,and determines the changes of ginsenosides and ginseng polysaccharides.Then,using dextran sodium sulfate-induced colitis mice as the model,the effect of processing methods on the efficacy of ginseng.The thesis first discusses the conversion of functional components of ginseng by the types of enzymes and processing conditions.The results show that the enzyme can promote the release of polysaccharides from ginseng and increase the ginsenoside Rd by about 5 times.Secondly,the paper used a variety of probiotics to treat ginseng,and found that probiotic fermentation can promote the release of different functional components in ginseng.The thesis also used enzyme-microbial composite processing technology to process ginseng,which showed that the content of Rd in the processed ginseng increased by about 10 times,the total saponin content also increased by 1.6times,and the number of probiotic bacteria in the product could reach 10~9CFU/ml.In order to examine the activity of ginseng lyophilized powder obtained by different processing methods,a mouse colitis model induced by dextran sodium sulfate was evaluated in this paper.Physiological changes in mice and intestinal injury results showed that compared with the oral control group of original ginseng powder,oral microbial treatment of ginseng and ginseng freeze-dried powder processed by microorganisms and enzymes can significantly alleviate the severity of colitis.The compound processed ginseng freeze-dried powder is more effective.Intestinal tissue damage assessment analysis by H&E staining method showed that compared with the oral original ginseng powder control group,oral microbial treatment of ginseng and ginseng freeze-dried powder processed by microorganisms and enzymes can effectively alleviate intestinal tissue damage caused by colitis.The effect of freeze-dried powder of ginseng compounded by microorganism and enzyme is more significant.The detection of inflammatory cytokines in intestinal tissues by enzyme-linked immunosorbent assay showed that oral microbial treatment of ginseng and ginseng compounded with microorganisms and enzymes can reduce the content of IL-4?IL-6?TNF-??IFN-?in the midgut tissue of mice with colitis.We further analyzed the 16S rRNA sequence of the mouse fecal genome,analyzed the abundance and composition of the intestinal flora of different groups of mice in different administration stages,and found that mice oral microbes treated ginseng and enzymes and microbes processed ginseng frozen The dry powder can effectively alleviate the change of intestinal flora caused by dextran sodium sulfate,and accelerate the recovery of intestinal flora towards health.Among them,the ginseng freeze-dried powder processed by microorganisms and enzymes is more effective.The method of compound processing of ginseng with enzymes and microorganisms not only improves the pharmacological composition of ginseng,but also incorporates probiotics with beneficial functions for the host.Oral enzyme and microbial compound processed ginseng lyophilized powder can reduce the severity of colitis in the mouse model and improve the change of the intestinal flora in the mouse model.This indicates that the method of compound processing of ginseng with enzymes and microorganisms is of great significance for improving the performance of ginseng.
