Study On The Regulation Of Estrogen And Its Receptor In Perimenopausal Model Rats By Zhuang Medicine Line Point Moxibustion Therapy

Objective To observe the effects of Zhuang medicine line point moxibustion therapy on estrogen and its related receptors in perimenopausal model rats,and to explore the effect mechanism of Zhuang medicine line point moxibustion therapy on estrogen and its related receptors in perimenopausal model rats.Methods Adult female SD rats were selected.The 4-VCD solvent was injected intraperitoneally for 15 days,and the 4-VCD dose was 80 mg/kg.Thirty successful rats were selected and randomly divided into 3 groups of 10 rats each.They were acupuncture group,acupuncture group and model group.Ten unmodeled rats were selected as normal control group.The normal group and the model group did not do any treatment and were reared in a conventional manner.In addition to the conventional breeding,the medicinal line group needed to be treated with medicinal point moxibustion.The acupuncture group was treated with acupuncture for 6courses.After the treatment,observe the general condition of each group of rats before and after the treatment.Samples were taken to determine the content of serum E₂,LH,FSH,and E₂in 1,3,5,and 6 cycles.ERm RNA protein and gene expression.Results 1.The results of serum E₂,LH,FSH and levels show that the E₂level of acupuncture group and moxibustion group is higher than that of the model group,and the LH and FSH levels are significantly lower than the model group;2.Periodic changes of serum E₂level:In each cycle,the E₂level of the point moxibustion group was lower than that of the acupuncture group in cycles 1,3,and from the cycle 5,the E₂level of the point moxibustion group began to be higher than that of the acupuncture group Cycle 6 reaches its peak;3.Western blot results suggest that the moxibustion group can increase the content of ER protein in the hypothalamus,pituitary and ovarian tissues and is higher than that in the acupuncture group;4.Immunohistochemical results showed that the moxibustion group could enhance the expression and distribution of ER protein in hypothalamus,pituitary and ovarian tissues,and the expression intensity was higher than that of the acupuncture group;5.RT-q PCR showed that the moxibustion group could increase the ERm RNA gene expression level in each tissue,and the expression level was higher than that in the acupuncture group.Conclusion 1.Zhuang Medicine Line Point Moxibustion Therapy can increase serum E₂levels,reduce LH,FSH,levels,and has a sequential nature;2.Zhuang medicine line point moxibustion therapy can increase the expression of ER protein and ERm RNA gene in hypothalamus,pituitary and ovarian tissue.Hormone level to achieve,its effect is better than acupuncture group.3.Zhuang medicine line point moxibustion therapy can improve ovarian function,relieve various symptoms of perimenopausal syndrome,and play a role in conditioning perimenopausal syndrome.
