Objective: By comparing the normal Sprague-Dawley?SD?rats and perimenopausal models of SD rats,normal gestational age women and perimenopausal women peripheral blood T lymphocyte subsets,CD4 +and CD8 + T lymphocyte connective protein(Connexin?P <0.05?,and the expression of IL-1,IL-2 and IL-6 in serum,and to investigate whether E2?estradiol?can inhibit the relationship between IL-1 and ILHormones caused by T lymphocyte subsets and proinflammatory cytokine release increased chronic inflammatory processes,thereby improving immune dysfunction caused by chronic inflammation.Methods: Animal Blood Sample Experiment: The experiment divided four-month-old female SD rats into control group,OVX group?perimenopausal period animal model group?and OVX+E2?estrogen replacement therapy group?.Flow Cytometry was applied to detect the ratio of T lymphocyte subsets in peripheral blood and the expression frequency of Cx40 and Cx43 on CD4+ and CD8+ T lymphocytes.Secondly,ELISA tech was applied to detect E2 difference?estradiol?between each group,expression of inflammatory factor IL-1,IL-2 and IL-6 of each group.Clinical Blood Sample Experiment: The objects were divided into childbearing age group and perimenopausal period group.Flow Cytometry was applied to detect the ratio of T lymphocyte subsets in peripheral blood of both groups and the expression frequency of Cx40 and Cx43 on CD4+ and CD8+ T lymphocytes;Meanwhile,ELISA tech was applied to detect expression of inflammatory factor IL-1,IL-2 and IL-6 of each group.Results: 1.E2 level of SD rats in OVX group was lower than that in the control group?P <0.05?.E2 level of OVX+E2 group was higher than that in OVX group?P <0.05?;2.CD4+CD25 + T lymphocytes of OVX group were higher than those in control group?P <0.05?;CD4+ CD25+ T lymphocytes in OVX+E2 Group were lower than those in OVX group?P <0.05?;CD4+/CD8+ in OVX group was lower than that in control group?P<0.05?;CD4+/CD8+ in OVX+E2 group was higher than that in control OVX group?P<0.05?;3.CD8+CX40 and CD4+CX43 in OVX group were obviously higher than that of Control Group?P<0.01?;CD8+CX40 and CD4+CX43 in OVX+E2 Group were lower than that of Control Group?P<0.01?;CD8+CX43of OVX+E2 Group was lower than both that of Control Group and that of OVX Group?P<0.01?.4.Rats' plasma IL-1? and IL-6 of OVX group were significantly higher than those in Control?P <0.01?,IL-2 was lower than that in control group?P <0.05?;IL-1?,IL-6?P <0.01?of OVX+E2 Group were lower than that in OVX group?P<0.01?;IL-2 was higher than that in OVX group?P<0.05?;5.Results of Pearson correlation analysis showed that,IL-1? and IL-6 of rats,were positively correlated with CD8+CX40,CD4+CX43 and CD8+CX43;IL-2 was negatively correlated with CD4+CX43;6.CD4+/CD8+ in the perimenopausal period group was lower than that of childbearing age group?P <0.05?;7.CD8+CX40,CD4+CX43CD8 + CX43 in peripheral blood of perimenopausal period group were higher than those of childbearing age group?P<0.05?.3.IL-2 and IL-6 concentration in peripheral blood of perimenopausal group were lower than those of childbearing age group?P<0.05?.Conclusion: The results above confirm that perimenopausal is a kind of chronic low degree inflammation,accompanied by abnormal ratio of T lymphocyte subsets and increase of proinflammatory cytokine release.Exogenous input of E2 can alleviate the damage caused by concomitant chronic inflammation of perimenopausal period above-mentioned,the possible mechanism of which is that E2 inhibits information communication between T lymphocytes by reducing the expression of Cx40 and Cx43 which constitute gap junction channels on T lymphocytes,thereby reducing the release of proinflammatory cytokines.
