The Development of Science and Technology and Management has promoted the progress and development of life science.Life science has become a new frontier of scientific research.Various new analytical and testing methods,technologies and equipment have been widely used in various fields of life science,such as revealing the mystery of life,studying the pathogenesis of diseases,and controlling diseases that endanger human health.Among many analytical and testing methods,biomass spectrum is considered as a universal method with high specificity and sensitivity and has been widely used.Especially in the field of proteomics and peptide omics,biomass spectrum seems to have become a synonym.But,more and more evidence that biological mass spectrometry technology in lipid omics and lipid analysis is also a very useful tool,because it has high sensitivity,less sample consumption,fast analysis speed,isolation and identification at the same time,can get rich in a few minutes of cell,tissue or fluid extract of biology information.In addition,the application of bio-mass spectrometry in the detection of small molecules of lipid metabolites is also restricted by other factors: in small molecule analysis,the traditional matrix will produce strong endogenous background peaks in the low molecular weight region.The spectrum is complex and difficult to distinguish,while suppressing the target analyte signal,interfering with the detection and identification of small molecule analytes.The commonly used organic small molecule matrix has not been able to meet various practical requirements in scientific research and clinical practice.A matrix containing a noble metal plasma material such as gold,silver,platinum or palladium provides unique surface plasmon resonance and heat carrier under laser illumination.Although there are many reports on the application of noble metal modified nanomaterials in biological mass spectrometry,most of the current work is used to develop a variety of precious metal nanoparticles to detect molecular weight smaller molecular weights such as a variety of amino acids and sugars.Little research has been done on the use of precious metal nanoparticles in the detection of small molecules in lipid metabolism.Therefore,the development of a special design of a new plasma matrix is of great significance for the application of lipid small molecule analysis in disease diagnosis.In this paper,a series of research work and development of new technologies and methods were carried out in combination with the surface functionalized magnetic nano-functional materials and lipid metabolism small molecule analysis in the hot spot of the detection of small molecules in lipid metabolism by bio-mass spectrometry.And apply the actual biological sample.In view of this,this thesis conducts research work around these contents,which is divided into three chapters.The main contents and results are summarized as follows:The first chapter outlines some basic concepts and typical applications of functional nanomaterials and biomass.Based on the brief introduction of functional nanomaterials and bio-mass spectrometry,the mechanism and principle of functional nanomaterials for bio-mass spectrometry are analyzed in a targeted manner.These mechanisms and principles are the main theoretical basis of this thesis.Finally,the topic and significance of this paper are explained.In the second chapter,a magnetic functional nanomaterial with core-shell structure was designed and fabricated.The core-shell structure,material selection and synthetic preparation technology of nanomaterials were expounded,and a new magnetic functional nanomaterial was synthesized based on this.Finally,the synthesized materials were characterized by various techniques and methods,and the performance data of nanomaterials were obtained.The stability and repeatability of nanomaterial synthesis experiments were verified.In the third chapter,the standard lipid samples were first tested using a series of synthetic nanomaterials.The test results were analyzed and the optimized nanomaterials were used for the testing of biolipid samples.Through experiments,it is fully demonstrated that our synthesized functional nanomaterials can be used as matrix materials.When bio-mass spectrometry is used to detect biolipid samples,sample preparation is simple,no complicated sample pretreatment process is required,and biological sample consumption is extremely low(nL).Expected research objectives.
