| Objective:This experiment aims to observe the effects and mechanism of RDN on lipid metabolism in simple obese rats.Methods:The SD rats were divided into two groups: normal diet group(NCD group,n=8)and high fat diet group(HFD group,n=22),rats of NCD group were used as control group and rats of HFD group were fed with high fat for 8 weeks to build up obese rats model.The obese rats(n=16)were randomly divided into two groups:SHAM group(n=8)and RDN group(n=8).Rats in SHAM group underwent sham operation and rats in RDN group underwent bilateral renal denervation(RDN).4weeks after operation we observed: 1)biochemical detection of serum TC,TG,HDL-C,LDL-C and serum FFA in rats of each group;2)determination of serum NE,renal artery HE staining by ELISA method to observe the changes of sympathetic nerve activity in order to judge the success of the operation;3)the levels of leptin and adiponectin in serum were measured by ELISA method;4)the expression of AMPK-PGC-1 α and its downstream molecular proteins in skeletal muscle were detected by Western-blot method.Results:After 8 weeks of high fat or normal diet feed,the weight and BMI of rats in HFD group were increased significantly compared with NCD group.4 weeks after RDN,the level of TG,TC,HDL-C,FFA in the RDN group decreased significantly(P<0.01)compared with the SHAM group.Compared with the SHAM group,the level of norepinephrine(NE)was significantly decreased in RDN group(P<0.01).Compared with NCD group,AMPK,p AMPK,PGC-1-α,UCP3,CPT-1 in SHAM group decreased significantly(P<0.05),however,after RDN these parameters get improved dramatically(P<0.05).Conclusion:RDN can improve lipid metabolism,which may be related to the regulation of leptin,adiponectin and AMPK-PGC-1 α pathway in skeletal muscle. |
